OBJECTIVE: To study the effect of Wulong-pills to Allergic rhinitis(AR) in Spleen Qi deficiency type and the mechanism of Wulong-pills in treating this disease from gene level. To establish the clinic treatment principle—tonify Spleen Qi and reinforce Surface—of AR through the Lab. Research from pathogenic physiologic changes .METHODS: Establishing rat's AR model or AR model in Spleen Qi deficiency type, 7 groups are involved which are the normal group, the Spleen Qi deficiency AR group, the Spleen Qi deficiency AR Chinese medicine group, the Spleen Qi deficiency AR Western medicine group, the AR group, the AR Chinese medicine group and the AR Western medicine group. Establish the rat's Spleen Qi deficiency modal with improper diet and over strain according to Li Dexin's report, AR modal according to An Yunfang's. After the model was established, Wulong-pills or Astemizole is given to the rat by enema starting from 23rd days to 29th days. During the treatment phase, the rats were stimulating by 0.5% OVA 1 hour after the medicine administrate, the author observed the symptoms especially nasal symptoms. After treatment, author choice 6 rats to apply the Light Microscope and Electron Microscope, to observe the pathogenic changes happened in the model rats, the others to apply RT-PCR to observe mRNA, such as IL-2 mRNA, IL-4 mRNA, IL-6 mRNA and IFN-γ mRNA. Meanwhile, pH of nasal cavity, histamine before and after treatment were examed.RESULTS: compare to the modal groups, Chinese medicine groups have significant functions of regulate the pH (P<0.01). Serum histamine of rats arise in the modal before treatment (P<0.01, compare to the normal group), and low down after treatment (P<0.01,compare with modal groups). All the treatment groups and the normal group have no differences in serum histamine. The rats in Chinese medicine groups have higher IL-2 mRNA and IFN-ymRNA express in nasal mucosa than the modal groups (p<0.01 or p<0.05), and lower IL-4mRNA, IL-6mRNA express in nasal mucosa( p<0.01 或 p<0.05). All the treatment groups and the normal group have no differences in the express of IL-2 mRNA , IFN-ymRNA , IL-4mRNA and IL-6mRNA in nasal mucosa(P>0.05). Spleen Qi deficiency AR and AR group have no significant differences in the express of IL-2 mRNA , IFN-γmRNA, IL-4mRNA and IL-6mRNA(P>0.05).CONCLUSION: Wulong-pills can release rat's experimental AR , such as antihistamine, inhabit the degrangulating of mast cell.
