Wulong Granule On Allergic Rhinitis In Spleen - Qi Deficiency Rat Mast Cell Signal Transduction Of Nasal Mucosa

OBJECTIVE: Allergic rhinitis (AR) is a common nasal disase, with high incidence. Wulong-pills is concluded from Pro. Xiong Dajing's long-clinic experience and has reliable treating effect on allergic rhinitis. We observe the effect of Wulong-pills on signal transportation of mast cell in nasal mucosa of AR rats modal, meanwhile, the change of signal transportation of mast cell in nasal mucosa of AR rats modal in Spleen Qi deficiency type and the regulation of Wulong-pills to this type.METHODS: Establishing rat's AR model or AR model in Spleen Qi deficiency type, 7 groups are involved which are the Normal group, the Spleen Qi deficiency AR group, the Spleen Qi deficiency AR Wulong-Pills group, the Spleen Qi deficiency AR Astemizole group, the AR group, the AR Wulong-Pills group and the AR Astemizole group. Establish the rat's Spleen Qi deficiency modal with improper diet and over strain according to Li Dexin's report, AR modal according to An Yunfang's. After the model was established, Wulong-pills or Astemizole is given to the rat by enema starting from 23 rd days to 29th days. During the treatment phase, the rats were stimulating by 0.5% VOA 1 hour after the medicine administration, the author observed the symptoms especially nasal symptoms. After treatment, 6 rats randomly choosed in each group to apply the Light Microscope and Electron Microscope, to observe the pathogenic changes happened in the model rats, at the mean time, they are used to do immunohita to watch the express of SP and ER~+. The others to apply RT-PCR to observe CK's mRNA, such as IL-2 mRNA, IL-3 mRNA, IL-10 mRNA and IL-12 mRNA. Meanwhile, pH of nasal cavity, histamine before and after treatment were examed.RESULTS: The results are as follow: Wulong-pills has significant functions of control the symptoms, low down the higher pH in nasal cavity and histamine in blood, reduce the pathologic changes of nasal mucosa, regulate nasal mast cells function, regulate signal transportation of mast cell through the followings: reduce the expression of SP and ER~+, enhance the expression of IL-2 mRNA and IL-12 mRNA in...
