As a traditional Chinese herb, Magnolia offininalis was firstly recorded in Shenlongbencaojing and can be found in every edition of Chinese pharmacopoeia. According to literature and reference book, the pharmaceutical part of magnolia is always the cortex in traditional herbal and the leaf of magnolia is never used. The cortex can't be peeled until the tree is 15years old. Cutting the trees is bad for protecting forest and ecology and will destroy the resource of Magnolia. Magnolia offininalis is widely planted in many places in large scale and the leaf of Magnolia offininalis is rich in resource and can be gathered every year. If the leaf of Magnolia offininalis can be utilized, it will be helpful to sustained utilization of the resource of medicinal material of magnolia and can bring great benefit to society and economy.Owing to the theory that different parts of a plant may have same or similar chemical component and have similar medical effect and clinical use, so we planed to do a range of experiments on acute toxicity, main medicinal effects and chemical components in order to investigate the medicinal value of the leaf of Magnolia.It's the first time to investigate the acute toxicity by the experiment of Maximal tolerated dose on mice. The result shows that the maximal tolerated dose for mice is 112g/kg, which is 747 times of human daily use, and there were no death and toxic reaction in 7days of observation.It's the first time to compare different extracts of leaf to that of cortex on pharmacodynamics. The results manifest that the low dose of petroleum ether extract of the leaf can lengthen delitescence time or reduce the frequency of tusses caused by strong ammonia solution; the low dose of Ether extracts of the leaf can improve the length percentage of the mixture propulsive effect of activated charcoal in small intestine; the high dose of Water extracts of leaf can make the mice lose the ability to coordinate muscular movement and resistance; The Water extract also have the some effect of bacteriostasis. All in all, the leaf and cortex of Magnolia have similar medicinal effect in main aspects.It's the first time to investigate the chemical components in leaf of Magnolia offininalis by systemic primary experiments. Hydroxybenzene, Alkaloid, Flavone, volatile oil are found in leaf, which are similar to chemical component in cortex..It's the first time to separate and confirm the chemical components in leaf of Magnolia offininalis .9 components have been separated and 7 of them have been confirmed by spectral analysis. They are palmitone, quercitrin, rutin, arachidic acid, 1-hexacosanol; β-sistosterol, daucosterol and all of them are separated from leaf of Magnolia for the first time.The contents of palmitone and quercetin have been determined for the first time. The results manifest that leaf contains about 0. 133~0.145% of palmitone and 0.147~0.153% of quercetin respectively.Above all, the leaf of Magnolia offininalis has some medicinal value. The results of these experiments have provided the scientific foundation for the further research and comprehensive utilization of resources of Magnolia offininalis.
