In our study, a rapid analyze method has been established to investigate the trace constituents in the bioactive extraction of medicinal plants by making full use of various mass spectrometric techniques. In order to investigate the relationship between the structural characterization and the mass spectrometric behavior of interested constituents, several natural products with the same structural skeleton were studied in detail by MS/MS technique. Some characteristic cleavage reactions which can reflect the connection between the fragmentation behavior and structural features were found and provide a valuable aid for determining the structures of related compounds. Although the mass spectrometry has been used widely in the study of the trace constituents extracted from medicinal plants, more structural information can be obtained and the analysis of the desired compounds in the mixtures can be simplified by the novel route presented in this paper. Under the direct of this method, the possible structure of the trace constituents in the extraction from Tylophora atrofolliculata and Rhizoma Coptids were inferred by using mass spectrometry (ESI-MS, FAB-MS, MS/MS, LC-MS and LC-MS/MS), and most of them were proved to be correct by LC-NMR techniques. Several novel and characteristic cleavage reactions, which can provide a valuable aid for determining the structures of related compounds, were observed during the study of the mass spectrometric behavior of phenanthriondolizidine alkaloids and protoberberine alkaloids.Many metal ions are present in the cell and are known to bind to DNA, such interactions are of great biological interactions. The mass spectrumetric behaviors of oligonucleotides are usually studied in negative ion mode. Here we report the mass spectrometry behavior of oligonucleotides and the complex ions with H~+, Na~+ and K~+ of them by using (+) ESI-MS/MS.
