The chemical consitituents and bioactivities of three Compositae and one Scrophulariaceae species were studies.One hundred and seven compounds were isolated,and twenty-one of them were new compounds.The Ligularia genus is genera of family Compositae tribe Senecioneae subtribe Senecioninae and perennial herbs,It consists of about 130 species in the world,with 111 species distributed mainly in northwest and southwest of China.More than 30 Ligularia species have long been used as folk medicine in China.Their roots,stems, leaves and flowers possess efficacies of antipyretic,relieving phlegm and relieving cough,invigorating the circulation of blood and soothing pain.Ligularia virgaurea (Maxim.)Mattf.is used as a traditional folk medicine for the treatment of stomachache and nausea.Continuing research for bioactive components from Ligularia species and the influence of different ecological environments on the chemical constituents of plants,we chose L.virgaurea collected in Gannan Tibetan Autonomous Region,Gansu Province.From the petroleum ether:diethyl ether: methanol(1:1:1)extract of the roots,thirtythree compounds were isolated.Their structures were elucidated by IR,UV,MS,1H,13C NMR(DEPT)and 2D NMR. Among these compounds,fifteen were new ones,including four novel asymmetric eremophilanes dimers,eight eremophilenolides and three noreremophilanes.The Sonchus genus is genera of family Compositae tribe Lactuceae subtribe Lactucinae and perennial herbs.More than 50 species were found widely in Europe, Africa and Asia.There are 8 species distributed in China,mainly in the Xinjiang province and southwestern mountainous region of China.Among them,5 Sonchus species have long been used as folk medicine for the treatment of fever,stasis and inflammation,as well as for detoxication and invigorating of blood circulation.From the methanol extract of the whole plant of Sonchus uliginosus which collected from Heyang county of Shanxi province,seventeen compounds were isolated.Their structures were elucidated as eudesmanolides,glycosides and phenylpropane derivatives etc.by spectral methods,of which three eudesmanolides and three phenylpropanes were new.The genus Ajania is genera of family compositae tribe Anthemideae subtribe Chrysantheminae.It consists of more than 30 species in the world.About 29 species are widely distributed in China besides southeast region.Many Ajania species have long been used as traditional Chinese medicine for the treatment of calming, antipyretic,relieving phlegm and relieving cough.From the methanol extract of the whole plant of Ajania tenuifolia which collected from Maqu county of Gansu province,thirtyfive compounds were isolated.Their structures were elucidated as terpenes,flavones and coumarins etc.by spectral methods.The genus Pedicularis is genera of family Scrophulariaceae.The genus Pedicularis consists of about 500 species,and 66%of these species growing in southwest of China.Many Pedicularis species are used in folk medicine as a tonic for the treatment of general debility,collapse,exhaustion,invigorating the circulation of blood,aid digestion,full of vitality,etc.From the ethanol extract of the whole plant of Pedicularis resupinata ssp.lasiophylla which collected from Taibai mountain of Shanxi province,twentytwo compounds were isolated.Their structures were elucidated as iridoid,lignan and phenylpropanoid glycosides etc.by spectral methods.Furthermore,the in vitro cytotoxic activities of some compounds against human hepatoma(SMMC-7721),human promyelocytic leukemia(HL-60),human cervical carcinoma(HeLa)and human hepatocyte(L-02)cells were carried out by sulforhodamine B(SRB)method.The 50%inhibitory concentration value(IC50) showed that compounds 1.1 and 1.3 exhibited moderate cytotoxic activity against HL-60 cells,and weak activity against SMMC-7721 and HeLa cells.Finally,the chemical components and bioactivities of the Ligularia species were reviewed in this thesis since 1998 to present.The results of this investigation would be expected to provide important scientific basis for the bioactive chemical constituents in traditional Chinese medicine, pharmacology,chemotaxonomy,screening new medicines,and utilization and development the natural resources of our country.
