Study On Treatment Of Extracts From Ailanthus Altissima On Mouse Ulcerous Colitis

The effect of the extracts from Ailanthus altissima was researched on ulcerous colitis. six kind of extracts were prepared by different extraction methods: extract of water, extract of water-alcohol deposit, extract of 95% ethyl alcohol, extract of 65% ethanol, extract of 45% ethanol, extract of 25% ethanol, and each extract constituent qualitative analysis as well as the medicine security experiment were carried on; experimental animals were divided into eight groups: the control group, the model group, and the different extracts groups stochastically, the ulcerative colitis(UC) models in mouse were made by 2,4-dinitrochloro-benzene (DNCB), each extract was filled the mouse by gavage, after 7 days, the function of each extract was appraised to the UC model mouse, MPO, PGE2, NO, NOS, SOD, MDA, GSH, GSSG, ADA and lysozyme were determined; the apoptosis of sensitized lymphocyte was researched during the home-nest process and information inheritance; the function of extracts to mouse exsomatize colon smooth muscles were observed.Experimental results: (1) Six kind of different extracts included different content of ingredient, and in the extract of water the phenol content was highest, moreover, extract of water-alcohol deposit includes the sugar, the polysaccharide and the glycoside materials. (2) The extract of water-alcohol deposit, extract of 45% ethyl alcohol, extract of 65% ethyl alcohol, extract of 25% ethyl alcohol could obviously promote body weight increasing, loss diarrhea, hematochezia of mouse, obviously recover from the mouse damaged colon organization, spleen and thymus, and could elevate the UC model mouse spleen index and the thymus index by varying degree, but to the extract of water and extract of 95% ethyl alcohol, the above function are not obvious. (3) the activity of MPO was suppressed, the content of PGE2 and the content of NO were reduceed, the activity of SOD was enhanced, MDA from Lipid peroxidation product in the UC model mouse colon organization was cleared, the value of GSH/GSSG was promoted, and content of ADA, lysozyme in blood serum, as well as ADA in spleen were lossed by varying degree. (4) The extracts the sensitized lymphocyte were caused to make the varying degree apoptosis during the home-nest process: The effect of extract of water-alcohol deposit was best, that of extracts of 65% and 45% ethyl alcohol were next best, and other extracts' function were not obvious. (5) The extracts had the direct stimulation to exsomatize colon smooth muscles: extracts of water and 95% ethyl alcohol had the greatly strengthened stimulation at the 200 times from original medicine, when density enlarging, the movement of the colon was suppressed obviously, the effect of extracts of water-alcohol deposit and 65% ethyl alcohol was remarkable, stable, there was no obvious difference among the different density, and the extracts of 45% and 25% ethyl alcohol could strengthen the contractive motion, but the movement rhythm and the intensity are unstable.It was concluded that in this study. (1) The extracts had certain treatment function to the mouse ulcerous colitis, but there was the difference among those extracts, the effect of extract of water-alcohol deposit was the best, but the effect of extract of water was the weakest. (2) The extrcts could promote organic function of immune organ, enhance system immunologic function, refine colon pathologic denaturation. (3) The extracts could adjust the redox reaction, eliminate many kinds of the oxygen free radical and the harmful products, and cause the redox reaction system to tend to the return to reduction state, thus the ulcerous colitis morbidity degree was alleviated, inflammation process was arrested. (4) The extracts could adjust the producing and metabolism of inflammatory materials, mediate the inflammation reaction, and promote inflammation retrogression. (5) The extracts could stimulate the lymphocyte apoptosis mechanism, cause the lymphocyte deactivation, suppress immune response occurrence, promote colon tissue inflammation restoration. (6) The extracts could strengthen the colon movement, improve colon nutrition condition, speed up the deleterious substance to discharge, and reduce the inflammation pathological degree. (7) The function of the extracts from Ailanthus altissima to mouse UC was a harmonious cooperation process with the multi-factors and multi-mechanism participation.
