Study On Sleep Improving Constituents Of Chinese Medicine Prescription Sinisan

Large clinical observation and empirical study indicate:chinesemedicine prescription sinisan has an unique curative effect on improvingsleep.In order to clarify active constituents of sedation function,activeconstituents of sedation function was studied.Freeze drying powder of sinisan was pilot studied by the method offingerprint spectrum and serum pharmacochemistry,compounds containingmensurations of Paeoniflorin,Naringin,Saikosaponin C,Glycyrrhetinic acidwere established.Extraction technology of decoction of sinisan wasestablished.These methods were Sinisan quality controlling standerdization;HPLC fingerprint spectrum of freeze drying powder of sinisan wasestablished at210nm,240nm wave length,the similarity of fingerprints wereestimated by correlation coefficient method.Meanwhile,the origins ofcompounds in the serum and cerebrospinal fluid were analyzed;Sinisansignificantly prolonged the sleep time of mice induced by pentobarbitalsodium freeze drying powder of Sinisan.The results indicated that:contents of the major ingredients in sinisanwere Paeoniflorin 2.55%,SaikosaponinA 2.01%,Naringin 3.17%,glycyrrhetinic acid 2.65%,it was in line with the national pharmacopoeia2005 standards.This experiment need add 10 times the volume of water,boiling 2 times 1 hour every time,and concentrated in a water bath at 70℃to0.4g (crude drug)/ml,using technology (-40℃refrigeration,-20℃securityfreeze 2h,-10℃security freeze 16h,20℃dry 5h,35℃secondary drying 2h.)Freeze drying powder of sinisan was freeze-type full,complex water-based,yellow,and porous mesh and water was less Than l%.Contents of the majoringredients in freeze drying powder of sinisan sinisan were Paeoniflorin notless than 2.74%,Saikosaponin A not less than 2.32 %,Naringin not less than3.31%.glycyrrhetinic acid not less than 2.70%,to ensure standardization ofthe experimental samples;There were fourteen peaks in the serum fingerprint spectium.Both compounds land 7 were found rooted in Fructus AurantiiImmaturus by the studv of the adscription of serum compound,They belong tothe constituents of Fructus Aurantii Immaturus and freeze drying powder ofsinisan which can be absorbed into blood directly;Compound 2 isSynephrine.Compounds 3,6 and 11 belong to the constituents of Radix EtRhizoma Glycyrrhizae and freeze drying powder of sinisan which can beabsorbed into blood directly;Compound 4 is Paeoniflorin;Compound 5 is anew substance which generated by the internal metabolizing;Compound 8 isthe metabolite of Radix Paeoniae Alba in vivo;Compound 9 is the metaboliteof Radix Et Rhizoma Glycyrrhizae in vivo;Compound 10 is the metabolite ofFructus Aurantii Immaturus in vivo.Compound 12 is Saikosaponin C;Compound 13 is a new substance which generated by the internalmetabolizing after adding Fructus Aurantii Immaturus and other medications;Compound 14 is Glycyrrhetinic acid.after confirming methanol methods toprepare cerebrospinal fluid samples and the time of gathering cerebrospinalfluid samples after oral administration of Sinisan seven days five hours asoptimal time,by the analysis of the compound in cerebrospinal fluid,wefound that there was no other compound absorbed into blood except for thePentobarbitalum Natricum.Moreover,the endogenous substance incerebrospinal fluid was increased obviously,Increasing peak area of theendogenous substance in cerebrospinal fluid,mixing standard preparation's(Paeoniflorin Naringin Saikosaponin C Glycyrrhetinic acid)is 3.2 timesSinisan's.The endogenous substance in cerebrospinal fluid wasn's5-hydroxvindole acetic acid,5-hydroxytryptamine,Norepinephrine,Dopamine.The component need be studied further.Results of thepharmacological experiment indicated that Sinisan significantly prolongedthe sleep time of mice induced by pentobarbital sodium.Finally.definiting active constituents of freeze drying powder of sinisanimproving sedation function,they are Paeoniflorin,Naringin,Saikosaponin Cand Glvcvrrhetinic acid.All the results provided a basic data for the further studies on effective constituents and the effecting mechanism of Sinisan.
