By using the method of Serum pharmaco-chemistry of Chinese Medicine, the analysis of constituents in blood after oral administration of Liu Wei Di Huang Wan have been carried out. Based on the establishment of the analyzing method of constituents in vitro, the HPLC fingerprint of the preparation and determination of loganin and paeonol in it were completed, the similarity of fingerprints were estimated by the vector angle method. Under the established HPLC condition, drug-containing serum fingerprint was set up and the constituents originated from the preparation and their metaboliates in the serum were identified by comparing fingerprint in vitro with that in vivo. As result, ten constituents have been detected in the blood after oral administration of Liu Wei Di Huang Wan, Compound I exists originally in Radix Rehmanniae, in blood it also derived from Fructus Coris and Rhizoma Alismatis; compound II ,IV and V belong to Fructus Corni, and compound VI and X are from Cortex Moutan's, compoundIII, VII ,VIII and IX are the metabolites of Paeonol contained in Cortex Moutan. All the results provide a basic data for the further studies on effective constituents and the effecting mechanism of Liu Wei Di Huang Wan.
