Objectives:To observe the natural progression of wound healing in rabbit maxillary sinus mucosa and to detect the expression of relevant cytokines.Methods:A rabbit wound-healing model was established in the maxillary sinus. At different times after injury, the sizes of maxillary ostia were recorded, and histological changes in the sinus mucosa were examined by means of hematoxylin and eosin (H&E) and Masson trichrome staining. Enzyme-linked immunosorbent assay (ELISA) was used to detect the concentrations of IL-6, IL-8, IL-10, and collagen types I and III.Results:A large amount of immature spindled collagen deposition was observed in the basal lamina and lamina propria of regenerative mucosa. IL-6 was increased significantly in regenerative mucosa after injury, especially at 14 days post wounding when compared with normal mucosa. IL-8 was also increased significantly in regenerative mucosa; however, IL-10 was decreased significantly when compared with normal mucosa. IL-6 and IL-8 showed a significant correlation with the expression level of collagen type I.Conclusion:Collagen deposition was increased in regenerative rabbit maxillary mucosa and the elevated collagen deposition during wound healing was associated with increased IL-6 and IL-8, and decreased IL-10. Objectives:To observe the role of IL-10 overexpression on the wound healing in a rabbit model of chronic bacterial sinusitis.Methods:A rabbit wound-healing model was established in the maxillary sinus with chronic bacterial sinusitis. A lentiviral vector (LV) expressing IL-10 and green fluorescent protein (GFP) (LV-IL-10) or GFP alone (LV-GFP) was injected at the wound site 3 days before wounding. Physiological saline was injected at the wound site 3 days before wounding as normal control. At 3 and 10 days post wounding, the sizes of maxillary ostia were recorded, and histological changes in the sinus mucosa were examined by means of hematoxylin and eosin (H&E) and Masson trichrome staining at 10 days post wounding. Real Time polymerase chain reaction (Real-time PCR) were used to measure the gene expression of IL-6, IL-10, TGF-β, and collagen types I and III. Enzyme-linked immunosorbent assay (ELISA) was used to detect the concentrations of IL-6, IL-8, IL-10, and collagen types I and III.Results:Both LV-IL-10 and LV-GFP were expressed in the wounds at 3 and 10 days post wounding. At 3 and 10 days, when compared with LV-GFP and physiological saline injection, LV-IL-10-treated wounds demonstrated decreased inflammation, collagen deposition and decreased quantities of proinflammatory mediators analyzed with statistically different levels of IL-6 and IL-8. However, there was no significant difference in the expression of TGF-P between three groups.Conclusion:Local application of lentvirus-mediated overexpression of IL-10 decreases the inflammatory response and collagen deposition post wounding in a rabbit maxillary of chronic sinusitis.
