Background The cerebral vessels diseases become one of the chief diseases which do harm to the human health. About 20 percents to 30 percents ischemia cerebral vessels diseases is relative to the carotid artery atherosclerosis stenosis. In order to study the carotid artery atherosclerosis.there must be a reliable animal model. Many experiments have confirmed that the artery atherosclerosis in swine is similar with human being, the mini-swine has the advantage of convenience to feeding, small volume and easy to management. We select the mini-swine and use the balloon catheter technique and highlipoid diet to establish the carotid artery atherosclerosis stenos model. Observe the markers such as the degree of stricture of artery, the kind of atheromatous plaque, and character of pathology, hemodynamics by the Doppler ultrasound and vessel pathology which verified that the animal model is according with the human carotid artery atherosclerosis stenosis.The interleukin-17 is a new predominantly proinflammatory cytokine in the past few years, IL-17 is a glycoprotein include 155 amino acid N-terminal signal peptide and secrete by the activated CD4+T-lymphocytes. IL-17 play several functions such as develop the inflammation, immune response, hemopoiesis by specific binding with the acceptor. The IL-17 has relative with several chronic inflammation and autoimmune disease. But the function of IL-17 in each stage of AS is controversial. Some studies found IL-17 can prevent the AS, there are also many researches find that the IL-17 can result in AS process. So the action of IL-17 in AS is controversial and more researches needed to proceed to reveal.The first part The establishment of the atherosclerosis stenosis animal model in carotid arteryObjective To create the animal model of the atherosclerosis stenosis in carotid artery.MethodWe selected 7 mini-swine to study, All the animal were fed with common feed for one week. The control group(one min-swine) continue to common feed, the experiment group(six min-swine) carried ultrasound carotid artery with the ketamine anesthesia, then carried the unilateral(right) carotid sacculus dilatation and fed with high lipid diet. Withdrawing the blood from the precaval vein to exanimate the triglyceride and cholesterol in the time of 2 weeks,4 weeks,8 weeks, and 12weeks. Observing the constrictive degree in damaged area in vessel. Put all of mini-swines to death, dissect the carotid artery, and observe the pathology changes in the damaged area.Result1 The ultrasound show that the of experiment animal artery have atherosclerosis apparently and have the atheromatous plaques and the vessel constriction. PSV、EDV is apparently higher than normal and vessel lacuna area deceased notably than normal. But the undamaged vessel is similar with the normal in vessel diameter.2. The triglyceride and cholesterol in 12 weeks is higher than before. It is the most highest in the 2 weeks.3. The pathology shows that the atherosclerosis of experiment groups is apparent. The intima incrassation and the new capillary vessel hyperplasia and more foam cells can be found. But the counter-lateral vessel can only find the intima incrassation lightly.ConclusionThe method can build the atherosclerosis stenosis animal model of mini-swine successfully by the sacculus damaged in carotid artery and high lipid diet. The ultrasound and pathology confirmed that the animal model is coincidence with the human being in each stage of carotid artery atherosclerosis. the animal model can be used in carrying out the experiment of carotid artery atherosclerosis stenosis reliably. The second part The expression of Interleukin-17 in the atherosclerosis stenosis animal modelObjective To observe the Il-17 expression in the intima, smooth muscle, atheromatous plaque in the animal model of the atherosclerosis stenosis and investigate the mechanism of IL-17 in AS.Method Apply the immunofluorescence staining technique to take photos and observe the expression of Il-17 in injured side or uninjured side arterial canal inside of the intima, middle smooth muscle, plaque, tunica extema to proceed the analysis and summary.Result The immunofluorescence showed that the IL-17 antibody aggregate in the injured artery obviously, and more gathered in the plaque but seldom in the intima and middle smooth muscle. There is no IL-17 aggregated in the uninjured artery.ConclusionIL-17 take part in the presses of AS and promote the formation of AS plaque, IL-17 play an important role as a proinflammatory cytokine in AS. But the mechanism of IL-17 is not revealed and need more researches to carry on.The Conclusion of the article1 The method that sacculus damaged in carotid artery and high lipid diet can build the carotid artery atherosclerosis stenosis animal model of mini-swine successfully. And the animal model is coincidence with the human being in each stage of carotid artery atherosclerosis verified by the ultrasound and pathology confirmed. the animal model can be used in carrying out the experiment of carotid artery atherosclerosis stenosis reliably.2 Verified the advantage of ultrasonic in detect the carotid artery atherosclerosis stenosis.3 The IL-17 participate in the process of atherosclerosis as a proinflammatory cytokine such as the formation and stability of plaque, and IL-17 play an important role in the AS, but the detail mechanism of action of IL-17 is unknown and need more research to reveal.
