Part I Comparison of two methods for establishing a model of experimental varicocele in ratsObjective To explore and compare two methods for establishing an animal model of experimental varicocele in rats.Methods90adolescent male Sprague-Dawley rats were randomly divided into three groups. In30rats (classic group, group A), this was achieved by dissecting the left renal vein and partial ligating it. In30rats (modified group, group B), in addition to the partial ligation of the renal vein, the communicating branch was fully ligated. In30rats (sham operation group, group C), the left renal vein was dissected, but not ligated. The operating time was examined in three groups. The diameters of the spermatic veins were analyzed respectively in4and8weeks later.Results the operating time of a rat was longer in group B than in group A and C(22.5±3.4vs.9.2±2.3vs.5±1.2min, P<0.05). The diameters of the left spermatic vein were (0.86±0.12)mm and (0.91±0.11)mm respectively in group A after4and8weeks, which was significantly more broadening than group C(P<0.01), and there was no statistical difference in group B(P>0.01). The model achievement ratio of4weeks in group A was lower than that of group B (70.59%vs.90.91%, P<0.05).Conclusion Both methods to establish the VC model in rats were feasible. To establishing model, the modified method has more operating difficulty and high achievement ratios in short term. Part II The effect and mechanism of hyperbaric oxygen on Nitric Oxide Synthase and Nitric Oxide in testis of varicocele ratsObjective To investigate the levels of nitric oxide synthase(NOS) and nitric oxide(NO) in testis of varicocele(VC) rats and to evaluate the potential role of hyperbaric oxygen(HBO).Methods50Sprague-Dawley(SD) male rats at age7weeks were randomly assigned into three groups.10rats were sham operation group(group A),40rats were created the experimental varicocele models by parial ligation of left renal vein, then were randomly divided into2groups, namely varicocele group(group B, n=20) and HBO administered after being induced varicocele model(group C, n=20). All testis were collected and the levels of NOS, NO were detected.Results The levels of NOS and NO in left testis of group B was significantly higher than that of group A and C(P<0.01), the levels of NOS and NO in right testis of3groups was no statistical difference (P>0.05).Conclusion NOS and NO over-expression in testis of varicocele rats may play an important role in causing aciesis, the HBO prevents testicular dysfunction probably by changing the levels of NOS and NO.
