| Objective:Study the effect and mechanism on component compatibility of Sini decoction to hypothroidism and it’s complications,in order to reveal the scientific connotation of component compatibility of Sini decoction to hypothyroidism.Method:1.The effect of component compatibility of Sini decoction to hypothy-roidism.The asdthenci splenonephro-yang of hypothyroidism model was induced by 0.1% PTU,and the level of thyroid hormone and thyroid pathological chan-ges were selected to study the impact of component compatibility of Sini Decoction to hypothyroidism thyroid fuction;choose the Bax,Bal,DA,NA, 5-HT to study the impact of component compatibility of Sini decoction to hypothyroidism induced brain damage;choose TG,TC and LDH-L to study the effect of component compatibility of Sini decoction to hypothyroidism induced lipid metabolism disorder;choose the serum BUN, CREA,Cys-C and kidney pathological changes to study the effect of com-ponent compatibility of Sini decoction to hypothyroidism induced renal inju-ry;and the myocardium Na+-K+-ATP enzyme and serum ANP,ET were dete-cted to study the the effect of component compatibility of Sini decoction to hypothyroidism induced cardiovascular damage. The general activity were also obsevated.2.The mechanism on component compatibility of Sini decoction to hypoth-yroidism.Choose the middle dose of component compatibility of Sini decoction which has the best effect to the hypothyroidism to study the mechanism.The level of TRHã€TSHã€T3 and T4were dectected to study the effect of component compatibility of Sini decoction to the hypothalamus-pitutiary-thyroid axis. The ACTH and CROT were dectected to study the effect of component compa-tibility of Sini decoction to the hypothalamus-pitutiary-adrenal axis.The level of TPO,TPO-Ab,Tg,Tg-Ab,TR-Ab,TAM-Ab and IL-1,TGF-β1were dectected to study the inmmune meachanism.The FQ-PCR were used to study the inful-ence of component compatibility of Sini decoction to NIS mRNA. The level of TTF-1,cAMP,PKA in thyroid were dectected to study the mocule mechanism of component compatibility of Sini decoction to hypothyroid-ism.3. The effect of component compatibility of Sini decoction to serum pro-teom in the hypothyroidism rat.The dimensional gel electrophoresis were used to separate the proteome, and analysis the changes of the protein expressions with PDquese 8.0.Results:1.The effect of component compatibility of Sini decoction to hypothy-roidism.Symptom:The 0.1% PTU can induced the asdthenci splenonephro-yang of hypothyroidism,the rat has some symptom such as dry and yellow hair,burnout and drowsiness,loose stools,et al.The symptom can improved after use the component compatibility of Sini decoction.The level of thyroid hormone and thyroid pathological changes:Compar-ed with the control group,the level of T3,T4 are reduced and TSH is increase-ed in model group (P<0.01);compared with model group, the level of T3,T4 are increased and TSH is reduced(P<0.05). Thyroid pathological changes sho-ws that the thyroid gland is obviously swelling and the tissue distribute in nodositas,vascular are increased,follicular epithelium hypertnphy,and some follicular are fused.Compared with the model group, pathological changes are reduced in the positive group and component compatibility of Sini decoction high and middle group.Hypothyroidism induced brain damage:The expression of Bax,Bal are no significant different in each group. Compared with the control group,the level of excitatory neurotransmitter NA and DA are decreaed and the inhibito-ry neuromitter 5-HT is increased(P<0.05) in the model group.In the drug grou- p the NA and DA are increased and 5-HT is decreased significantly.Hypothyroidism induced renal injury:Compared with the control group, the level of BUN,CREA and Cys-C increased significantly,and have some pathological changes; Compared with the model group, the level of BUN, CREA and Cys-C increased significantly decreased (P<0.05),Pathological changes are reduced in the positive group and component compatibility of Sini decoction group.Hypothyroidism induced hyperlipemia:Compared with the control group, the level of TG,TC,LDH-L are increased significantly; Compared with the mo-del group, the level of TG,TC,LDH-L are decreased in the positive group and component compatibility of Sini decoction group(P<0.05).Hypothyroidism induced cardiovascular injury:Compared with the contr-ol group,the level of ANP, ET and Na+-K+-ATP enzyme are decreased signif-icantly(P<0.01); Compared with the model group, the level of ANP and ET are increased(P<0.05),but the level of Na+-K+-ATP enzyme has no significant different between the drug group and model group.2.The mechanism on component compatibility of Sini decoction to hypothyroidism.Neuroendocrine regulation:Compared with the control group,the level of T3,T4 are reduced and TSH,TRH is increased in model group (P<0.01);Com-pared with the model group, the level of T3,T4 are increased and TSH, TRH are decreased significantly(P<0.01). Compared with the control group,the lev-el of ACTH and CROT are increased significantly, compared with the model group, the level of ACTH and CROT are decreased(P<0.05). It shows that the component compatibility of Sini decoction can regulate the hypothalamus-pitutiary-adrenal axis and hypothalamus-pitutiary-thyroid axis.Immune regulation:Compared with the control group,the level of TPO,TPO-Ab,TAM-Ab,TGF-β1 are reduced significantly(P<0.01), the level of Tg,Tg-Ab,TR-Ab,IL-1 are increased significantly(P<0.01). Compared with the model group, the level of TPO,TPO-Ab,TAM-Ab,TGF-β1 increased signi-ficantly,and the level of Tg,Tg-Ab,TR-Ab,IL-1 are reduced.It shows that the component compatibility of Sini decoction can regulate the immune system.Gene regulation:Compared with the control group,the expression of the NISmRNA are reduced in the model group; Compared with the control group and model group, the expression of the NISmRNA are increased significantlyin the component compatibility of Sini decoction group(P<0.01).Cytokine regulation:Compared with the control group, the level of TTF-1,PKA,cAMP are significantly increased in model group(P<0.01), the level of TTF-1 increased significantly in component compatibility of Sini decoction group; Compared with the model group,the level of TTF-1,PKA reduced, the level of cAMP are no difference between the model group and component compatibility of Sini decoction group.3. The effect of component compatibility of Sini decoction to serum proteom in the hypothyroidism rat.Compared with the control group,there are 34 spots of protein expression increased and 67 decreased, component compatibility of Sini decoction can increase 14 spots of protein expression and decrease 7 spots of protein expres-sion.Conclusion:1. The component compatibility of Sini decoction has good effect to hypothyroidism and hypothyroidism induced brain injury, renal injury Hyperlipemia,cardiovascular injury.2. The component compatibility of Sini decoction can regulate the neuroendocrine systerm,immune systerma, gene expression and cytokine in order to cure the hypothyroidism.3. The component compatibility of Sini decoction can affect the serum protein expression of hypothyroidism rat. |
