Multiple sclerosis (multiple sclerosis, MS) is considered the destruction of myelin sheath caused by own immune effector T cells, It is a central nervous system disease which can make the person be disabled. Shugan Jianpi Gusui prescription is a foundation prescription used to treatment MS in relief period. It already showed a good curative effect on the clinical good and it has a bright prospect of research the prescription. The further research of Shugan Jianpi Gusui prescription is of practical significance. The objective of this research is further understanding MS pathological mechanism on bioinformatics. The mechanism of Shugan Jianpi Gusui prescription treating MS was analyzed on the level of molecular biology. Then it provided molecular biology substratum for Shugan Jianpi Gusui prescription used to treating MS. Further, the optimization of Shugan Jianpi Gusui prescription was guided.The effect of Chinese herbs on T subsets of multiple sclerosis: an Analysis by Association RulesObjective:Analyzed the effect of Chinese herbs on peripheral blood T subsets of multiple sclerosis, to provide evidence for optimizing prescription.Methods:Organized the information which had been inputted in China academy of traditional Chinese medicine Xiyuan hospital cases acquisition system of MS including outpatient clinic and ward. Using Spss 17.0 and Aprior module of Spss Climentine 12.0, analyzed of the relationship between traditional Chinese medicine (TCM) herbs and T cell subsets.Results:① Radix bupleuri, radix paeoniae alba, fructus aurantii, atractylodes and radix glycyrrhizae are the most commonly used herbs combinations. Radix aconiti lateralis preparata and rhizoma smilacis glabrae are often added.② Radix aconiti lateralis preparata and Thl cells decreased had relevance principle (Radix aconiti lateralis preparata=> Thl cells decreased, Confidence 83.78%, Support Level 36.26% and Th1 cells decreased=> Radix aconiti lateralis preparata, Confidence 71.26%, Support Level 36.26%).③ The prescription of radix bupleuri, radix paeoniae alba, bitter orange, atractylodes, radix glycyrrhizae and Radix aconiti lateralis preparata could reduce peripheral blood Thl subset of MS(P=0.0000) and could rise Th2 subset (P=0.0003).Conclusion:the herbs combination of radix bupleuri, radix paeoniae alba, fructus aurantii, atractylodes, radix glycyrrhizae and Radix aconiti lateralis preparata could prevent relapse of MS by reducing peripheral blood Thl and enhancing Th2, especially Radix aconiti lateralis preparata playing an important role.The bioinformatics analysis and coding proteins interaction network of multiple sclerosis expression difference genesThe objective is further understanding the MS pathological mechanism and looking for MS potentially interventional targets. In this research we choose dataset form GEO (Gene Expression Omnibus) database which is contain microarray data of MS whole blood and normal humen whole blood. Used unpaired t-test to filter differentially expressed genes of MS, implemented GO analysis and Reactome pathway analysis and protein-protein interaction network. We screened 267 differentially expressed genes in MS whole blood in which 192 are up-regulated and 75 are down-regulated. GO analysis indicated that up-regulated genes are mainly enriched in immune defense, immune regulation, and T cell-mediated immune process and down-regulated are mainly enriched in concentrated in the nucleic acid/nucleotide metabolism, mRNA transport and adjustment process. Reactome pathway analysis showed that up-regulated genes are mainly on immune related pathways and down-regulated are centralized on chromosome painting, assembly elated pathways. Conclusion:MS immune system change is result from transformation of mitochondria and cells energy metabolism, and mitochondrial dysfunction and cells energy metabolism related proteins is members of MS potential target protein.The research on pharmacology network of Shugan Jianpi Gusui Prescription treatment of multiple sclerosisObjective:Exploring Shugan Jianpi Gusui prescription’s effective components influence MS different expression gene encoding proteins comparing to the normal. Method:MS related target protein as a medium, Shugan Jianpi Gusui prescription’s all components and protein targets were docked by the computer molecular simulation docking technology. The mechanism of Shugan Jianpi Gusui prescription treated MS were analyzed on the level of molecular biology.Results:None compounds of Baizhu docking with MS target proteins were included in Drug-Target interaction network. There were 23 compounds of Shugan Jianpi Gusui prescription having highly network degree, and 10 compounds of Shugan Jianpi Gusui prescription strongly binding to MS target proteins. 1/3 key proteins are targets proteins in targets proteins and different genes encoding proteins interaction network.Conclusion:Shugan Jianpi Gusui prescription can be optimized for basic prescription being composed of gancao, chaihu, zhiqiao, shaoyao, fangfeng, fuzi. Glycyphyllin and Isopaeoniflorin not only bind to multiple target proteins but also bind at high chemical energy, so these 2 compounds can be as candidate of treatment MS compounds in the future researches. Shugan Jianpi Gusui prescription could regulate MS different genes encoding proteins by affect targets proteins.
