Objective Network pharmacology,molecular docking technology and in vitro experiments were used to study the potential mechanism of action of Fangfeng decoction in the treatment of osteoarthritis(OA),which laid a more reliable theoretical foundation for the subsequent treatment of OA.Methods The main active ingredients and targets of Fangfeng decoction were screened through the pharmacology database of traditional Chinese medicine system and Uniprot database,and the main targets of OA were found through Gene Cards database,Pharm Gkb database,OMIM database,TTD database and Drug Bank database.Cytoscape software was used to construct a "Fangfeng decoction active ingredient-OA core target" interaction network model.Subsequently,KEGG enrichment analysis was performed on the intersecting targets.The main components and core targets obtained were verified by molecular docking technology,and the core chemical components were verified by in vitro experiments.CCK-8,q PCR and Western blot were used to investigate the effects of different concentrations of quercetin on the expression levels of related proteins and m RNA in human osteoarthritis chondrocytes induced by IL-1β.Results The core active ingredients of Fangfeng decoction in the treatment of OA were quercetin,luteolin,kaempferol,baicalein,baicalein,etc.,and the core targets were STAT3,TNF,AKT1,ESR1,TP53,etc.Molecular docking technology verified that the active components of Fangfeng decoction could effectively bind to OA related target proteins.The biological pathway by which Fangfeng decoction regulates OA mainly acts on lipid and arteriosclerosis,AGE-RAGE signaling pathway in diabetic complications,fluid shear stress and atherosclerosis.Its molecular functions are mainly nuclear receptor activity,ligand-activated transcription factor activity,transcription co-regulatory binding and so on.Molecular docking results shows that the core components of Fangfeng decoction has good affinity with key targets of osteoarthritis.The results of cell experiments showed that quercetin with different concentrations of IL-1 β-induced C28/I2 cells could significantly inhibit the expression of matrix metalloproteinase in chondrocytes,inhibit cartilage degradation,and effectively protect chondrocytes.Conclusion Fangfeng Decoction treats OA through multi-component,multi-target and multi-pathway mechanism,which provides a basis for the clinical development and utilization of Fangfeng decoction in the intervention of OA.
