Purpose: To establish the rat model with chronic obstructive pulmonary disease, set up the blank group, model group, and ginseng extract high, medium and low dose group, test group rats blood CD3 +, CD4 +, CD8 + expression rate and serum IgG, IgA, IgM level and IL- 18, the activity of TNF- a, analysis of IL- 18, TNF- a mRNA gene expression, NF and kB signaling pathways in the pathogenesis of COPD; Proven ginseng extract on the immune function of COPD rats and inhibit its airway inflammation is the ways to adjust by NF- kB.Method: Will be 60 SD adult male rats were randomly divided into 6 groups, each group of 10, only model group, ginseng extract respectively high, medium and low dose group and positive medicine(dexamethasone) in the control group. By observing the group overall situation, the lung tissue of rats, HE staining pathological evaluation of COPD animal model of rats and ginseng extract of pathological changes of the COPD rats and the influence of the level of inflammatory cells; After using MASSON staining to observe each group in the rat model of collagen content, the thickness of the smooth muscle, etc.; Application of image analysis system to determine the COPD rats model of thickness, the inner diameter of the trachea, bronchi smooth muscle calculation index, the collagen fiber area of bronchial smooth muscle; Shipped in blood flow cytometry instrument measured the expression of CD3 +, CD4+, CD8 + ratio, and calculate the CD4 + / CD8 + ratio; By ELISA kit in the determination of serum IgG, IgA, IgM level and IL- 18, the activity of TNF- a, Liquid nitrogen frozen ground tissue mRNA were extracted with trizol, reverse transcription, to design primers for real- time PCR amplification, based on internal controls, observation group IL- 18 lung tissue, TNF- a mRNA gene expression; Application kit to extract tissue protein, the use of protein imprinting(WB, Western blotting) method, the determination of each lung tissue expression of NF- kB and I- kB.Result:1. The COPD rats model reduced airway inflammation, collagen content, area and bronchial smooth muscle of the collagen fiber index improved, airway remodeling, the effect is similar to or superior to dexamethasone, especially in the high, middle dose group of ginseng extract is preferred;3. Ginseng extract can regulate cytokine levels, and reduce the COPD rats serum IgG,IgA, IgM level and IL- 18, the activity of TNF- a, thus improve the COPD rats cellular and humoral immune function, reduce the number of the COPD acute;4. Different doses of ginseng extract on the COPD rats model IL- 18, concentration of TNF- a inflammatory markers and IL- 18, TNF- amRNA has certain the influence of the expression of lung tissue concentration-response relationship, especially in the high, middle dose group of ginseng extract is preferred, ginseng extract can inhibit airway inflammation,improve immunity, enhance its ability to disease resistance;5. Ginseng extract inhibit airway inflammation, improve the pathological damage of lung tissue COPD, action mechanism of the prevention and treatment of COPD by NF and kB signaling pathways, raised the expression of I- kB.Conclusion: This experiment successfully copied COPD animal model of rats, this study targeted stronger; By analyzing relevant data, can be confirmed by regulating serum, ginseng extract IgG, IgA, IgM level and IL- 18, the activity of TNF- a, enhance the immunity of the copd experimental rats, inhibit airway inflammation, improve airway remodeling, its mechanism through the NF- kB signal pathway inhibition of airway inflammation of copd rats, regulate immune function; At the same time through cut inflammation factors and adjust the immune function of copd rats.
