This experimental research is aiming to investigate the inhibitory effect of MIL60on CNV and compare it with other commonly used agents using animal models.Part â… :Krypton Ion Laser-induced BN Rat Models of CNVObjectives:to establish BN rat models of CNV using krypton ion laser techniques and investigate the development of CNV and cautions in the establishment of models. Methods:35BN rats (aged6-8weeks) were selected and evenly randomized into7groups.6observation groups were subjected to krypton ion laser-induced CNV and the control group were not. Subsequently, the fundus fluorescein angiography (FFA) results at different time points. Results:after being subjected to krypton ion laser-induced CNV, the BN rat models presented with genesis of CNV at7d, rapid growth of CNV at14d, peak at21d-28d, stabilized structure after28d. Conclusion:krypton ion laser techniques are characterized by short time and high success rate in inducing CNV animal model.Part â…¡:Electroretinogram of MIL60-treated BN Rat Models of CNVObjectives:to investigate the effect of MIL60on the electroretinogram (ERG) of krypton ion laser-induced BN rat models of CNV. Methods:The retinal electrophysiological waves were recorded for normal BN rats and krypton ion laser-induced BN rat models of CNV using ERG. Results:Krypton ion laserr-induced BN rat models of CNV presented with significant decrease in a-and b-waves in Scot-ERG, which suggested that the internal and external layers of retinal cells had been damaged. Conclusion:intravitreal administration with MIL60can improve the repair of internal layers of retinal cells in CNV.Part â…¢:The Experimental Research on the Inhibitory Effect of MIL60Objectives:to investigate the inhibitory effect of MIL60on the proliferation and migration of Huvec and RPE cells. Methods:MMT colorimetric method and Transwell were used in evaluating the the inhibitory effect of MIL60on the proliferation and migration of Huvec and RPE cells. Results:krypton laser-induced BN rat models of CNV presented with proliferation and migration of Huvec and RPE cells, which were inhibited by MIL60in a concentration-dependent way. Conclusion: VEGF plays an important part in the proliferation and migration of Huvec and RPE cells and its activities can be inhibited by MIL60.Part IV:Comparison between MIL60and Other Agents in Inhibition of CNVObjectives:to compare the inhibitory effect on CNV between MIL60and other agents intravitreally administered and primarily evaluate the efficacy of MIL60in the treatment of CNV. Methods:55BN rats (aged6-8weeks) were selected and randomized into7groups, respectively MIL60group (n=10), Lucentis group (n=10), Avastin group (n=10), triamcinolone group (n=10), BSS group (n=10) and the control group (n=10).7d after induction, all groups but the control group were intravitreally administered with the corresponding agents. FFA and the maximum thickness of CNV were observed at different time points. Results:56d after induction, MIL60group, Avastin group and Lucentis group showed no statistical difference in different indexes and significantly transcended the triamcinolone group and the BSS group. Conclusion:MIL60has a strong and persistent inhibitory effect on the development of CNV, which is similar to Avastin and Lucentis.
