Objective:According to the "preventing disease from exacerbating" theory of TCM,this article is to study the effects of Jiawei Huangqin decoction on intestinal inflammation as well as the tumor and to explore the relationship with the regulation of IL-6/STAT3signaling pathway at the whole, the molecular and the cellular levels. It clarifies the relationship between IL-6/STAT3signaling pathway and chronic inflammation as well as cancerization to prevent the cancerization of inflammatory bowel disease (IBD) and eventually reduce the incidence of colorectal cancer. It tries to bring about new research ideas for prevention of colorectal cancer and explore new prevention methods.Methods: Experiment one:70rats were randomly divided into normal controls group, model group, Jiawei Huangqin decoction group, mesalazine group, Huangqin decoction group, purslane group. There were10rats for each group. The models of TNBS induced colitis rats were established according to the methods from literatures. Pathology section of the colon tissuewas obtained.The content of serum IL-6was detected by adopting radioimm-unoassay method. MPO was detected by utilizing immunehistochemical staining. sIL-6Ra and gp130were detected by using Enzyme-linked Immunoassay. STAT3and IL-6mRNA were detected by adopting the method of RT-PCR.Experiment two:75mice were randomly divided into normal controls group, model group, Huangqin decoction group, Jiawei Huangqin decoction group, purslane group. There were15mice for each group. The models of colitis in mice relating to colo (CAC) were copied according to the methods from literatures.Histopathology obervation, apoptosis index, ELISA detecting MPO and TNF-a level were detected.Experiment three:50mice were randomly divided into normal controls group, model group, cyclophosphamid group, Huangqin decoction group, Jiawei Huangqin decoction group. There were10mice for each group. The tumor-bearing mice models with implanted sarcoma180(S180) were established and the mice colonic carcinoma (CoCon-26) were cultivated. IR was observed through the anti-tumor effect in vivo. The proliferation of T-lymph cells was detected. The anti-tumor effect in vitro was examined. The function of the mice colonic carcinoma (CoCon-26) was detected.Findings:1.Jiawei Huangqin decoction can improve food intake and defecation of inflammatory bowel disease (IBD) and relieve intestinal mucosa edema in rats, which has the remarkable difference in comparison with model group, Huangqin decoction group, and purslane group. In terms of serum IL-6, intestinal tissue MPO, serum sIL-6Ra, the content of gpl30, STAT3,and the expressive level of IL-6mRNA, Jiawei Huangqin decoction compared with themodelgroup on astatistically significant difference (P<0.01) and with Huangqin decoction group, and purslane group on a statistically significant difference (P<0.05). Huangqin decoction group, and purslane group have no statistically significant (P>0.05).2.Jiawei Huangqin decoction can relieve intestinal mucosa edema in mice with inflammatory bowel disease (IBD) and reduce body’s inflame-matory response, which has the remarkable difference in comparison with model group, Huangqin decoction group, and purslane group. In terms of apoptosis index, MPO, the influence of TNF-a, Jiawei Huangqin decoction compared with the model group on a statistically significant difference (P<0.01) and with Huangqin decoction group, and purslane group on a statistically significant difference (P<0.05). Huangqin decoction group, and purslane group have no statistically significant (P>0.05).3.Jiawei Huangqin decoction can accelerate the proliferation of T-lymph cells and inhibit the growth of tumor in tumor-bearing mice as well as the proliferation ability of colonic carcinoma (CoCon-26). Jiawei Huangqin decoction compared with the model statistically group on a Significant difference (P<0.01) and with Huangqin decoction group, purslane group on astatistically significant difference (P<0.05). Cytoxan group has no statistically significant (P>0.05).
