Part One Protein P27 and Osteopontin(OPN) expression in prostate cancer organizations and the correlation analysisResearch background Prostate cancer(PCa) is a disease which has serious impact on human health. Itis one of the most commonly seen tumorin men patient.Although in the past few decades prostate cancer diagnosis and treatment technology has made great progress, but its incidence and recurrence rate is still high.According to the statistics of one of America’s scientific research institutions’ autopsy of some elder men patients(more than 50 years of age),the prevalence of PCa is 10% ~ 45%,The older the patient’s age, the higher the risk. The reason is mainly due to the pathogenesis of prostate cancer is not clear so far, and the related research progress is also relatively lagging behind.Convenient and stable experimental animal model is the important means and tools for the study of prostate cancer clinical and basic research, it is of great significance for exploring the prostate cancerincidence and the prevention and control mechanism. Epidemiological studies have found that age and ethnic factors, genetic factors, diet, hormones and other risk factors are not only the risk factors for the attack of prostate cancer, but also closely related to the evolution of it.The causes forprostate cancer in patients with familial aggregation lie in the following: genetic predisposition, exposed to a common environmental factors or just caused by a high incidence of accident.The study shows that the incidence of prostate cancer in double egg twins is significantly lower than that of monozygotic twins,which indicates that hereditary factor occupies a very important factor in the etiology of prostate cancer. Prostate cancer has a connection with the patient’s lifestyle, especially the patient’s food that is rich in fat, meat and milk.The fatty acid peroxide in food can produce a kind of carcinogenic damage in the process of peroxide.This substance involved in fatty acid peroxide enzyme AMACR excessive expression in the tissue of prostate cancer, androgen levels in patients also play a critical role in the process ofthe development of prostate and the progress of prostate cancer.In animal experiments, testosterone and dihydrotestosterone can induce prostate cancer. So the pathogenesis of prostate cancer is an extremely complex process, which is a result of multiple factors.At present, some pathogenesis is still not clear.Purpose of research Through this study, it aims to explore and establish the expression of protein P27 and osteopontin(OPN) in related prostate cancer tissue, and it also includes the para-carcinoma tissue. All the research is based on statistical analysis.Research method Select diagnosed patients with Pcaafter prostate biopsy or surgical pathology from my hospital, Sep. 2007-Dec.2012.By usingimmunohistochemical methods, observethe expression of P27, OPN inthe tissue of patients with prostate cancer. In the same way,detect the expression of P27, OPN in the corresponding tissue adjacent to carcinoma biopsy.Then analyze its relevance to the clinical stage, tumor differentiation degree, and the tumor metastasis pathological factors.Research results Protein P27 expression was significantly higher in the tissue adjacent to carcinoma than in the tissue of prostate cancer, which proves this kind of protein’s main negative impact on the cell cycle regulation.It adjusts the extracellular negative colonization signal enhancement, conveys this information to the cell, thus functioningas a kind of tumor suppressor genes. So the cell proliferation ability can be shown through its high or low expression. If the body cells are affected by the bad information over a given period of time, P27 protein expression level is characterized by declining trendat this time, cell cycle of conversion thus will be completed.Within the organization there is no other way to prevent tumor cells’ proliferation, division, finally to the malignant lesions, and the OPN expressed in metastatic prostate cancer tissue is higher, which also proves OPN’s important role in the process of tumor occurrence, development and its transfer: it can induce cell proliferation and transformation, weaken immune cells’ effects on tumor killing; promote the formation of tumor blood vessels and the destruction of local membrane structure;Mediate the adhesion of cells and extracellular matrix;By inhibiting cell apoptosis and helping tumor cells to evade the immune system’s recognition and destruction, it can promote the invasion and metastasis of tumor cells, P27 and OPN expression in prostate tissue showed a negative correlation.Conclusion 1. By using immunohistochemical methods,the study observes protein P27, OPN expression in patients’ prostate cancer tissue.The finding shows the two express negative correlation. 2. Protein P27, osteopontin(OPN) have statistical significance in cancer clinical stage, differentiation, whether the tumor metastasis and other factors.Part Two An analysis on the expression of protein P27, Survivin and osteopontin(OPN) in prostate cancer tissueResearch objective Through this study, explore and establish the expression of protein P27, Survivin and osteopontin(OPN) in related prostate cancer tissue, and the expression of P27 protein Survivin and osteopontin(OPN) in the tissue adjacent to carcinoma, by the application of statistical analysis.Research method Select diagnosed patients with Pcaafter prostate biopsy or surgical pathology from the first hospital affiliated to Suzhou University, Sep. 2007-Dec.2012.By usingimmunohistochemical methods, observethe expression of P27, Survivin and OPN inthe tissue of patients with prostate cancer. The expression of Protein P27,protein Survivin and Osteopontin(OPN) have statistical significancein cancer clinical stage, differentiation, whether the tumor metastasis and other factorsPart Three The analysis of the correlation PSA and between expression of Clusterin and Survivin in prostate cancer tissueObjective:Bymeans of detecting the analysis of the correlation PSA and between expression of Clusterin and Survivin in prostate cancer tissue we want to investigate significance oftherapy to PCa.Methods Immunohistochemistry was employed to measure the expression of Clusterin and Survivin35 cases of prostatic cancer and 30 cases benign prostatic hyperplasia and PSA in plasma.Results In prostatic cancer tissue, the positive expression rates of Clusterin and Survivin were 74.2%(26/35)and 54.3%(19 /35). In benign prostatic hyperplasia tissue, the positiveexp- ression rates of Clusterin and Survivin were 0(0 /30)、0(0 /30),The positive expression of Clusterin and survivin between prostatic cancer and benignprostatic hyperplasia were significantly different(P < 0.05).The positive expression of Clusterin and survivin were related with the histologicgrade, clinical stageandmetastasis of prostatic cancer and PSA(P<0.05). The expression of survivin was not a reverse correlation with Clusterin expression in prostatic cancer(P>0.001)Conclusion Thedetection expression of Clusterin and Survivin and PSA in PCa may help to offer a new mode for the prevention and treatment of prostatic cancer.
