Curcuma comosa Roxb.also named as "Wan chak motluk" in Tailand,is from the family of Zingiberaceae(ginger family),which distributes throughout tropical and subtropical regions of the world,especially Indonesia,Thailand and Malaysia.The rhizome of C.comosa is used as an anti-inflammatory agent in Thailand.The previous bioactivity study on the methanolic extract of C.comosa showed an obvious antiinflammation effect.In order to clarify the constituents of C.comosa,the research on the chemical constituents of it was carried on. Silical gel,ODS,HPLC chromatography columns,and IR,UV,NMR,MS,CD, along with chemical derivation were employed to separate and identify structures of C.comosa.The structures of these compounds were established as follows:consisting of 31 sesquiterpenes:they were comosoneⅠ(1), comosoneⅡ(2),dimethoxycurcumenone(3),comosoneⅢ(4),curcumenone (5),comosol(6),curcolonol(7),curzerenone(8),isofuranodienone(9), furanodienone(10),1(10)Z,4Z-furanodiene-6-one(11),germacrone(12), 13-hydroxygermacrone(13),germacrone-1(10),4-diepoxide(14), dehydrocurdione(15),neocurdione(16),curdione(17), 7αr-hydroxyneocurdione(18),7β-hydroxycurdione(19),zederone(20), zederone epoxide(21),glechomanolide(22),alismol(23),isoprocurcumenol (24),procurcumenol(25),aerugidiol(26),zedoalactone B(27),zedoarondiol (28),isozedoarondiol(29),alismoxide(30),curcumadione(31);6 phenolic compounds:they were 4-(3,5-dihydroxyhexyl) phenol(32), 4-hydroxybenzaldehyde(33),4-hydroxy-3-methoxybenzaldehyde(34), 4-(4-hydroxyphenyl) butan-2-one(35),(+)-rhododendrol(36),4, 6-dihydroxy-2-O-(β-D-glucopyranosyl) acetophenone(37);seven monoterpenes and their glycosides:they were comosoxide A(38),comosoxide B(39),comososide(40),2,2,6-trimethyl-1-oxaspiro[2,5]oct-5-en-4-ol(41), 1-hydroxy-α,α,4-trimethyl-3-cyclohexene-1-methanol(42),6-hydroxy-3 (1-hydroxy-1-methylethyl)-6-methyl-2-cyclohexen-1-one(43),(1S,2S, 4R)-2-hydroxy-1,8-cineoleβ-D-Glucopyranoside(44);twelve diarylheptanoids: they were(3S’,5S’)-3,5-diacetoxy-1-(3, 4-dihydroxyphenyl)-7-(4-hydroxyphenyl) heptane(45),(3S’,5S’)-3, 5-dihydroxy-1-(3,4-dihydroxyphenyl)-7-(4-hydroxyphenyl) heptane(46),(3R’, 5S’)-3,5-dihydroxy-1-(3,4-dihydroxyphenyl)-7-(4-hydroxyphenyl) heptane(47), (3S’,5S’)-3-acetoxy,5-hydroxy-1,7-bis(4-hydroxyphenyl) heptane(48),(3S’, 5S’)-3,5-diacetoxy-1,7-bis(4-hydroxyphenyl) heptane(49),(3R,5S)-m-3, 5-dihydroxy-1,7-bis(3,4-dihydroxyphenyl) heptane(50),(3R’,5S’)-3, 5-dihydroxy-1 -(3-hydroxy-4-methoxyphenyl)-7-(3,4-dihydroxyphenyl) heptane (51),(3S’,5S’)-3,5-diaceoxy-1,7-bis(3,4-dihydroxyphenyl) heptane(52),(3S’, 5S’)-3,5-dihydroxy-1,7-bis(3,4-dihydroxyphenyl) heptane(53),(3R’,5S’)(3, 5-dihydroxy-1 -(3,5-dimethoxy-4-hydroxyphenyl)-7-(4-hydroxyphenyl) heptane (54),(5S)-5-hydroxy-1,7-bis(4’-hydroxyphenyl)-3-heptanone(55),1, 7-bis(4-hydroxyphenyl) hepta 4E,6E-dien-3-one(56).Among them,sixteen were identified as novel compounds,i.e.compound 1-4,6,32,38-40,45, 47-51,54;compound 41 was identified as a novel natural compound;five compounds were isolated from the genus of Curcuma for the first time,i.e. compound 11,23,42-44;and all the known compounds except 37 were isolated from this plant for the first time.
