TCM Syndrome Type, Syndrome Analysis Of Endometrial Lesions And Molecular Mechanism Of Qingrejiedu Intervention

Objective:A retrospective research was hold in cases with endometrial hyperplasia diseases and EC,to analyze and summarize the distribution of TCM syndromes.To investigate the expression of "autophagy" in endometrial lesions by measuring the expression of LC3 in pathological tissues of endometrial lesions.The effects of method of qingrejiedu "Lianhua Tang" extract on the proliferation and apoptosis of human endometrial carcinoma cell line HEC-1A cells and Ishikawa cells were studied experimentally,To study the mechanism of "Lianhua Decoction"extract on autophagy of humanendometrial expression of "autophagy" in endometrial lesions was investigated.According to the results of LC3 test,the relationship between autophagy and TCM syndromes and TCM syndromes was discussed.MTS method was used to determine the inhibitory effect of "Lianhua Tang" extract on proliferation of endometrial cancer cell lines HEC-1A and Ishikawa in vitro.Effects of "Lianhua Decoction" Extract on Apoptosis and Cycling of Endometrial Carcinoma Cell Line HEC-1A and Ishikawa Cells by Flow Cytometry.Effect of "Lianhua Decoction " Extract on the Invasion Ability of Endometrial Carcinoma Cell Line HEC-1A and Ishikawa Cells by TranswellWestern blot was used to detect the effect of Lianhua Decoction extract on the expression of LC3 protein in endometrial carcinoma cells.Real time PCR was used to detect the expression of LC3B gene in endometrial carcinoma cells.Immunofluorescence staining was used to observe the effect of" Lianhua Decoction"extract on autophagy protein LC3 in endometrial carcinoma cellsEffects of Lianhua Decoction extract on autophagy of endometrial carcinoma cells by transmission electron microscopy.Result:The most common TCM syndromes of endometrial hyperplasia and endometrial cancer cases is Qi and blood stasis,kidney deficiency and kidney yin deficiency,Blood stasis,qi deficiency,heat(Fire),Yin deficiency,wet,hot and humid is frequency in endometrial cancer.LC3 in the diagnosis of traditional Chinese medicine in the distribution from high to low,followed by broken off,menorrhagia,intermittent bleeding,Zhengjia,uterine bleeding and menstrual extension.The value of LC3 in endometrial carcinoma was higher than that in endometrial hyperplasia group,which was statistically significant.Different TCM syndromes LC3 measured values are different,the highest value in the determination of kidney deficiency.Compared with normal group treatment with n-butanol extract form Lianhua decoction significantly increased cell apoptosis rate,decreased cell proliferational rate(P<0.01)."N-butyl alcohol extract from Lianhua decoction" has significant inhibitory proliferation effect in different concentration and timeon 2 kinds of endometrial cancer cells.Between 24 hours,48 hours groups,HEC-1Acellon 1.25mg、0.625mg,0.3125mg cell inhibition rate had significant difference(P<0.01);Ishikawa cell on 5 mg/mL,2.5 mg/mL,1.25 mg/mL,0.625 mg/mL,0.3125 mg/ml,0.15625 mg/ml cell inhibition rate had significant difference(P<0.01)(P<0.01);HEC-1 a cells,Ishikawa cells on 10 mg and 5 mg,0.625 mg,0.078125 mg cell inhibition rate had significant difference in 24 hours(P<0.01)."Compared with normal group,the apoptosis rate of N-butyl alcohol extract from Lianhua decoction on HEC-1A and Ishikawa cells was significantly higher.Compared with normal group,The HEC-1A cell:high dose of 0.4 mg/mL,middle dose group0.2 mg/mL,and low dose of 0.1 mg/mL,cell apoptosis rate had significant difference(P<0.01);Ishikawa cells:on high dose of 1.0 mg/mL,,middle dose group0.5 mg/mL and low dose group 0.25 mg/mL cell apoptosis rate have significant difference(P<0.05).The n-butanol extract form Lianhua decoction can effectively inhibit the cell proliferation and induce cell apoptosis.Conclusion:The most common TCM syndromes of endometrial hyperplasia and endometrial cancer cases is Qi and blood stasis,kidney deficiency and kidney yin deficiency,Blood stasis,qi deficiency,heat(Fire),Yin deficiency,wet,hot and humid is frequency in endometrial cancer.The n-butanol extract form Lianhua decoction can effectively inhibit the cell proliferation and induce cell apoptosis.
