Electrophoresis,as an advanced detection technology,is widely used in biochemical analysis,medical detection,food safety monitoring and material sample preparation.In particular,electrophoresis can combine or fuse with other technologies to create unique results.The application of moving raction boundary theory in biochemical separation and analysis has spawned the electrophoretic titration(ET)technology.The establishment of acid-base ET model has provided the foundation for protein content detection and the structural information determination such as protein acid and alkaline residue base on the technology.As a quantitative analysis method,electrophoresis titration technology can convert chemical signals into visualized distance signals and realize rapid detection and analysis without relying on complex large-scale instruments or equipment.However,there is still necessary to use the difference of boundary migration distance to study the quantitative analysis of sample content.The chip based electrophoretic titration technology has potential to apply to different scene but with great challenge.Besides,it is also of great significance to develop a stable and reliable instrument platform to realize the automatic detection of electrophoresis titration for the practical application of large sample detection.To solve the issues mengtioned above,first,the nanoparticles ET model for ligands detection was developed base on the moving reaction boundary,a numerical model of nanoparticles ET was use to verify the boundary migration,the neutralization reaction could convert to visual diatance and measure conviently.The results have good consistency compared with NMR results.Second,the temparture effection in ET investigated based on a capillary electrophoresis(CE)heat dissioation model,the joule heat and temperature of channel cross section calculated,a numerical model developed by considering the effects of temperature to simulate the ET boundary migration,the numerical results have good consistency to experimental results.A double inner standard analysis model developed to realize the rapid quantitative analysis of protein content in different conditions.Third,we developed a new ET equipment: 1)a new large size ET device designed and manufactured.Considering the bubble problem in ET,the internal structure of the electrode chamber optimized to realize the bubble diversion and exclusion,providing a stable operating environment for the titration experiment.In addition,the replacement of the electrophoretic titrator without leakage of electrode buffer realized through the structural optimization,and improved the safety of the experiment.2)An automatic boundary detection system was developed based on imaging measurement technology.To avoid multifarious manul post-processing of a mass data and meet the rapid analysis of difference samples.non-contact automatic measurement carried out on the moving reaction boundary through optical imaging technology.
