The micro-channel network on the electrophoresis chip is manufactured by the technology of Micro Opto Electro Mechanical System on the substrate of glass, fused silica, PDMS etc. Sample separation is performed in the micro-channel and detected by Opticcal or Chemical methods. Electrophoresis chip is a multidisciplinary technology, It became a new platform in the field of analytical chemistry, especially in the life science, drug discovery and medicine.Electrophoresis chip technology is one of leading edge technologies of modern analysis science. The performance of every micro fluid device which constructs the whole electrophoresis chip system is very significant, especially to optimization of the whole system. Although electrophoresis chip is widely applied, the mechanism of microfluidic on this kind of chip is far from well known. The research on the liquid fluid characteristic of micro fluid device can provide instruction to the chip design and analysis of its function. Simulation to the design can effectively reduce the cost and improve efficiency. Meanwhile, it is an important way to study micro fluid. In this dissertation study of fluid field on two kinds of chips is presented, including simulation of the liquid fluid field on the chips with simulation software Coventor Ware , ecperimental validation and optimization of chip design based on the conclusion.The topics of this dissertation are presented below:Discussed the joule heat of electrophoresis chip, a comparison between electrophoresis chip and traditional CE system on joule heat, In order to know the effect of the Joule Heating in micro channels, the governing equations for the microfluidic are numerically simulated by the finite difference method. An optimization design was made for the chip. Numerical simulation is done to evaluate the influence of these factors, and experiments were also performed to validate the simulation result. Both theoretical analysis and experiment results show that the optimization design in our existing device can achieve the desired results.With Coventor Ware softmare, mathematical model is built up to evaluate the influence to the flow of different parameters, setup numerical model based on inrensive study of electrophoresis chip and the software package (Coventor Ware) to improve the original chip design. low voltage electrophoresis chip has been used to protein separation experiments; the results showed that it has met the need for protein separation protein BSA and Lys, and the results of experiments accord with the theory computation.The dissertation shows that it is can effective way to study the fluid characteristic on chips with numerical simulation. Numerical simulation method has some advantages such as convenience and accuracy, as well as breaking through the restriction of the experiment method. It is valuable to the design of fluid path and optimization of chip system. It is believed that numerical simulation method will be adopted in a large amount in the electrophoresis chip research, and surely promotes the development of electrophoresis chip research.
