Purpose This study was to investigate the changes of cytokines in aqueous humor in patients with different types of glaucoma,and study the inhibitory effect of salidroside on glaucoma-related cytokines and trabecular meshwork fibrosis.It provides new insights for clinical treatment of glaucoma.Materials and methods 1.By collecting aqueous humor samples in different types of glaucoma patients,the levels of TGFβ2,SFRP1,Gremlin and other cytokines in the aqueous humor were assayed by ELISA,and the relationship between the levels of them and intraocular pressure were analyzed.2.Human primary trabecular meshwork cells was cultured and identifed.To establish a fibrosis model of trabecular meshwork cell,trabecular meshwork cells were induced by dexamethasone.Intervening with salidroside,the expression of cytokines such as TGFβ2,SFRP1,and Gremlin1 and the extracellular matrix proteins such as FN,COL-IV,and LN were detected.This was used to evaluate the effects of salidroside on inhibiting the fibrosis of trabecular meshwork.3.Ocular hypertensive mouse model was made by using adenovirus of TGFβ2 overexpression vector.We explored the effect of salidroside for therapeutic ocular hypertensive mouse model.Results 1.The levels of TGFβ2 and SFRP1 in the aqueous humor of the acute angle-closure glaucoma(AACG)with high intraocular pressure group were significantly higher than those in the AACG with normal intraocular pressure group.The levels of SFRP1 in POAG with normal intraocular pressure group was significantly higher than that in POAG with high intraocular pressure group.2.Dexamethasone could increase the levels of extracellular matrix proteins such as FN,COL-IV,and LN and TGFβ2,Gremlin1,SFRP1 and other cytokines in trabecular meshwork cells.After intervention of salidroside,the expression levels of extracellular matrix proteins and cytokines was reduced obviously.3.Salidroside can significantly reduce the intraocular pressure in ocular hypertension model mice constructed with TGFβ2 overexpressing adenoviral vectors.Conclusion 1.The expression of TGFβ2 and SFRP1 in the aqueous humor of patients with acute angle-closure glaucoma was positively correlated with IOP.2.Salidroside could inhibit the fibrosis of trabecular meshwork cell and reduced the expression of cytokines.3.Salidroside reduced intraocular pressure in the ocular hypertensive model and provides an experimental basis for salidroside treatment of primary open angle glaucoma. |
