Objective To explore the mechanism of Shanghuo caused by Korea Red Ginseng through AMPK’s function in regulating energy metabolism.Methods(1)The rat model of excessive Shanghuo was established by feeding high,medium and low dosage Korea red ginseng’s Concentrated decoction for 15 days.The level of TSH and 17-OHCS was detected by Elisa assay.Protein and gene expression of AMPK、PGC-1α、NRF1 was verified by Western blot and real-time PCR.Na+-K+-ATP enzyme、Ca2+-Mg2+-ATP enzyme and SDH were detected by kits.(2)Healthy volunteers take Korea red ginseng until they were diagnosed Shanghuo.Blood regular test,hepatic and renal function were tested between the control group and the Shanghuo group.Gene expression of AMPK、PGC-1α、NRF1 was verified by real-time PCR.SDH was detected by kit.The gas chromatography-mass spectrometry was used to test the plasma metabolites between two groups.The significant changes in metabolites and associated metabolic pathways were excavated by using OSC-PLS DA analysis and t-test method,in order to explain the corresponding changes in the body under Shanghuo and its relation to AMPK.Results(1)Compared with the control group,expression of TSH and 17-OHCS were significantly increased in low,medium and high dosage group(p<0.05).Protein expression of AMPK,PGC-1αand NRF1 were significantly up-regulated in medium and high dosage group(p<0.05);m RNA expression of AMPK,PGC-1αand NRF1 were significantly up-regulated in low and high dosage group(p<0.05);Acivity of Na+-K+-ATP enzyme、Ca2+-Mg2+-ATP enzyme and SDH were significantly increased in medium and high dosage group(p<0.05).(2)In the clinical research,people suffer from Shanghuo after taking Korea red ginseng for average 13.9±2.72 days.Compared to the control group,m RNA expression of AMPK,PGC-1αand NRF1 as well as SDH were significantly up-regulated(p<0.05),glucose,xylose,palmitic acid,stearic acid,valine,serine were significantly up-regulatedand cholesterol was significantly down-regulated in the Shanghuo group(p<0.05).Conclusion(1)In the animal research and clinical research,vigorous status of energy metabolism was tested in Shanghuo caused by red ginseng,correlated with activation of AMPK,leading to upregulation of energy metabilisom related factors as well as accelerated tricarboxylic acid cycle.(2)In the clinical research,the differential metabolites was tested in Shanghuo people,which reflected enhanced glucose metabism,amino acid metabolism and fat metabolism,correlated with AMPK’s function in regulating energy metabolism,indicating vigorous status of energy metabolism in the body. |
