Insomnia is called insomnia in traditional Chinese medicine,which is caused by mental malnutrition or restlessness,and its main manifestation is that it can not get normal sleep.Aromatic Chinese medicine has unique advantages in the treatment of insomnia,depression and other chronic sub-health diseases.It can play a role in improving mood and synergistic treatment in the prevention and prognosis of diseases.Aromatic plant essential oils can play a good sedative and hypnotic role through nasal administration and sniffing.This topic is guided by the theory of aromatherapy of traditional Chinese medicine(TCM),and the research is carried out under the key theory and technology of aromatherapy of TCM,which is Aroma Road,Aroma Effect,Aroma Titigation-Aroma Stabilization-Aroma Evaluation.Aiming at the deficiency of both the heart and the spleen and the insomnia symptoms possibly induced by the deficiency of both the heart and the spleen,the research on the aromatic essential oil of traditional Chinese medicine is carried out,so as to provide a theoretical basis for the research on the treatment and improvement of the insomnia symptoms of the insufficiency of both heart and spleen by aromatherapy,explain the action mechanism,develop innovative health products and promote the development of the health industry of traditional Chinese medicine aromatherapy.1.Data Mining of Aromatic Traditional Chinese Medicine in the Treatment of Insomnia with High Frequency DrugsStarting from aromatic traditional Chinese medicine,the ancient classical prescriptions for insomnia were collected as comprehensively as possible,the core high-frequency aromatic traditional Chinese medicine was refined by data mining,and the potential medication rules were analyzed and summarized to find out the prescriptions for insomnia and the frequency of drug use in ancient books and modern Chinese patent medicines.To screen out the most frequently used drug types and the most frequently used drug combination in the treatment of insomnia.According to the screening results to determine the angelica,Atractylodes macrocephala as the treatment of insomnia model drug study.To provide the basis for exploring a better treatment plan and developing related aromatic products.2.Analysis of Volatile Oil Components in Angelica Sinensis and Atractylodes Macrocephala and Optimization of Extraction Process and Modeling Analysis of Extraction KineticsThe traditional steam distillation method was used to extract volatile oil from Angelica sinensis and Atractylodes macrocephala,and the results showed that there were little differences in oil yield,chemical components and relative content between the two methods.The extraction technology of volatile oil from Angelica sinensis and Atractylodes macrocephala was optimized by central composite design.Finally,the extraction process of angelica,Atractylodes macrocephala extraction kinetic model fitting,according to the angelica volatile oil yield evaporation loss is not obvious,using a number of formulas were fitted,found that the diffusion model of spherical particle fitting effect is better.In view of the evaporation loss of volatile oil from Atractylodes macrocephala Koidz,the calculation formula of oil yield with time was derived,and the fitting results were good.It lays the foundation for the follow-up research and the development of related products.3.Network Pharmacology Analysis of Angelica and Atractylodes Volatile Oil in the Treatment of Heart and Spleen Deficiency InsomniaThe interaction network of component-target-disease was constructed by network pharmacology.The material basis and the mechanism of the angelica and the largehead Atractylodes rhizome for treating insomnia with deficiency of both the heart and the spleen are analyzed,related target proteins of effective components in angelica-largehead Atractylodes rhizome volatile oil are screened out according to the component analysis result of the angelica and the largehead rhizome,an active component-action target network diagram is constructed,Go enrichment analysis and KEGG pathway enrichment analysis were performed.The results showed that angelica-atractylodes volatile oil mainly in the role of including acetylcholine receptor,adrenoreceptor,noradrenoreceptor,dopamine receptor,5-hydroxytryptamine receptor,endothelin receptor regulation to achieve the treatment of heart and spleen deficiency insomnia.The components of volatile oil from Angelica sinensis and Atractylodes macrocephala are mainly involved in the interaction pathway of neuroactive ligand-receptor.Corresponding verification is carried out through molecular docking.To provide a theoretical basis for the mechanism of Angelica-Atractylodes volatile oil in the treatment of insomnia of heart and spleen deficiency type.4.Synergistic effect of volatile oil from Angelica sinensis and Atractylodes macrocephala on pentobarbital sodium and sleep in miceBy study a mouse sleep model induced by that pentobarbital sodium and stud the influence of the medicament on indexes such as the sleep rate,the sleep latency time,the sleep duration and the like of the mouse according to the righting reflex condition of the mouse,It has certain sedative and tranquilizing effects.The middle dose group and the low dose group of Angelica-Atractylodes macrocephala volatile oil showed better sedative and tranquilizing effect.5.Study on the Effect of Volatile Oil of Angelica and Atractylodes Macrocephala on Heart and Spleen Deficiency RatWeight-loading swimming method combined with propranolol and reserpine was used to establish the syndrome model of deficiency of both heart and spleen in rats,and the insomnia model caused by asthenic fatigue and mental dystrophy was established.After modeling,the model rats appeared weight loss,reduced food intake,heart,brain blood supply decreased.Intestinal propulsion hyperfunction and increase of behavioral activity index and other symptoms.After administration angelica macrocephala water extract high dose group and angelica evaluated.6.Acute Toxicity of Volatile Oil from Angelica Sinensis and Atractylodes MacrocephalaThe acute toxicity of volatile oil from Angelica sinensis and Atractylodes macrocephala Koidz was studied by nasal administration after aromatherapy,and the toxic effect on the body and the toxic reaction of related symptoms and histopathological examination of main related organs were observed.To evaluate the safety of volatile oil from Angelica sinensis and Atractylodes macrocephala by intranasal administration.The organ index results showed that: Angelica-Atractylodes volatile oil group mice heart,liver and other organ index compared with the blank group were no significant difference.Brain,spleen,lung,kidney organ index compared with the blank group have a certain degree of significant difference.It is suggested that the volatile oil of Angelica sinensis and Atractylodes macrocephala has obvious effect on the index of some organs of mice.The damage of brain and lung tissue was the most serious,the damage of liver and kidney tissue was the second,and the damage of nasal mucosa tissue was lighter.7.Study on the suitability of volatile oil of Angelica sinensis and Atractylodes macrocephala for intranasal administrationThe volatile oils of Angelica sinensis and Atractylodes macrocephala were prepared into microemulsion,microemulsion gel and microcapsule according to the characteristic of slow release for the treatment of chronic diseases such as insomnia.The effects of several suitable dosage forms on the stability,absorption and bioavailability of the volatile oils were compared and investigated through in vitro release and stability indicators.Results The prepared formulations had good controlled release and improved the stability of volatile oil to a certain extent.In summary,through data mining to identify Angelica,Atractylodes macrocephala as the treatment of insomnia high frequency of the classic prescription drug pair.The volatile oil prepared under the optimized extraction conditions was stable and the extraction rate was high;The results of network pharmacology research showed that Angelica sinensis and Atractylodes macrocephala volatile oil could treat insomnia of heart and spleen deficiency type mainly by regulating the interaction of nerve active ligand-receptor;The results of pharmacodynamics experiments showed that the volatile oil from Angelicae Sinensis and Atractylodes Macrocephala Koidz had certain sedative and tranquilizing effects on the mouse sleep model induced by pentobarbital sodium,and also had certain therapeutic effects on the animal model of insomnia of deficiency of both heart and spleen.The mechanism of the effect may be related to the main targets of 5-hydroxytryptamine(5-HT),dopamine(DA)and gamma-aminobutyric acid(GABA),etc;The compound volatile oil preparation prepared by the new preparation technology is stable and controllable,and it provides theoretical and technical basis for the secondary development of traditional prescriptions.
