| Objective: To study the effects of components and group distribution of Atractylodes Rhizome and Bran fried Atractylodes macrocephalae L.on the improvement of gastrointestinal function and regulation of intestinal flora in rats with spleen deficiency and dampness syndrome,and hope to improve the gastrointestinal function and intestinal flora.The index of each indicator establishes the evaluation system of the components and groups of Bran fried Atractylodes macrocephalae L.to treat the spleen deficiency,and provides a theoretical basis for the synergistic promotion or inhibition of the group distribution.Methods: 1.According to the principle of atractylation of Atractylodes,the components of the same batch,Atractylodes Rhizome and Bran fried Atractylodes macrocephalae L.were extracted and separated,and the main components were determined by GC-MS,HPLC,phenol-sulfuric acid method.2.Establish a rat model of spleen deficiency and dampness syndrome by using the method of bitter cold and diarrhea and the method of hunger and dysfunction.The volatile oil component,the lactone component,the polysaccharide component and the lactone volatile oil component and the lactone plus polysaccharide component obtained by extracting Atractylodes Rhizome and Bran fried Atractylodes macrocephalae L.The rats in the rats with distress were treated with intragastric administration.The normal group,the rhubarb model group and the positive drug group were established for comparison.The changes of body weight and behavioral activities were observed and recorded.3.On the 2nd day after the last treatment,the rats were anesthetized and the abdominal aorta was taken for blood.The contents of GAS,VIP and SS in the serum of each group were determined.4.The faeces and cecal contents of rats were collected during anatomy.The fecal characteristics of rats in each group were recorded.The changes of intestinal flora in each group were observed by microscopy and selective culture.Results: 1.Extract isolated Atractylodes and Bran fried essential oil component,a lactone component and a polysaccharide fraction was found by measuring the content of essential oil components and Aractylone of the content significantly higher Bran fried Atractylodes;inner Atractylodes The content of ester components and main components of atractylenolide I,atractylenolide II and atractylenolide III was significantly lower than that of Bran Fried Atractylodes;the content of polysaccharides in Atractylodes macrocephalae L.was lower than that of Bran Fried Atractylodes.2.After the 14 th day of modeling,the weight of the 14 groups of rats in the model group decreased compared with the normal control group,the food intake was reduced by half,and accompanied by obvious abdominal swelling,the stool appeared,the pile collapsed,the blinking was less,Weight loss,rectal prolapse and other symptoms.After the start of treatment,the diet and body weight of each group of rats increased,and other symptoms were relieved to varying degrees.After 7 days of treatment,the rhubarb model group was still swollen in the abdomen compared with the other groups,but the food intake and body weight increased slightly during the modeling period.Compared with the rhubarb model group,the normal control group,the positive group,the Bran Fried Atractylodes macrocephalae suspension group,the Bran Fried Atractylodes lactone group,the atractylenolide plus polysaccharide group,the Bran atractylenolide plus polysaccharide group after treatment There was a significant increase in body weight(P<0.01 or P<0.05),and the weight of the other groups was higher than that of the model group but there was no significant difference.Compared with the rhubarb model group,the GAS content in the serum of the Bran Fried Atractylodes lactone plus polysaccharide group,Bran Fried Atractylodes lactone group,positive group and Bran Fried Atractylodes macrocephalae suspension group was significantly increased(P<0.05).The Bran Fried Atractylodes lactone plus polysaccharide group and Bran Fried Atractylodes lactone group were the most significant(P<0.01),and the GAS content in the serum of other groups also increased to varying degrees,but none of them were significant.Sexual difference;the content of SS in the serum of the Bran Fried Atractylodes lactone plus polysaccharide group,Bran Fried Atractylodes lactone group,Bran Fried Atractylodes polysaccharide group,positive group and Bran Fried Atractylodes macrocephalae suspension group were significantly increased(P <0.05),the VIP content was significantly decreased(P<0.05),and the decrease of VIP content was the most significant in the Bran AAtractylodes lactone plus polysaccharide group(P<0.01).The other groups had different degrees of change,but none of them were significant.Sexual differences.4.Rats in the rhubarb model group had loose stools,and the feces were thin and not formed.The proportion of Bifidobacteria and Lactobacillus colonies was the largest,and the proportion of Enterococcus and E.coli colonies was the smallest.The feces of normal rats and Bran fried The feces of Atractylodes macrocephalae polysaccharide and Bran Fried Atractylodes lactone plus polysaccharide group were close to spheroidal.The proportion of Bifidobacterium and Lactobacillus colonies was the smallest in the three groups,and the proportion of Enterococcus and E.coli colonies was the largest.The feces of the atractylenolide group were viscous and obviously wet,and there was no significant difference between the Bifidobacterium,Lactobacillus,Enterococcus and E.coli colonies and the rheum officinale model group.Conclusion: Compared with Atractylodes,the volatile oil component of Bran Fried Atractylodes Rhizome decreased,and the content of lactones and polysaccharides increased.The treatment of spleen deficiency and dampness syndrome rats with Bran atractylenolide and polysaccharide component was the most effective,which could improve and regulate the spleen deficiency and dampness syndrome.The gastrointestinal function of the mouse and the intestinal flora confirmed the synergistic effect of the group distribution. |
