In pan Dunhuang, whose position was special and critical, there were precious biodiversity resources. Recent years, the population increased rapidly and ecological environment deteriorated constantly in there. As a result, the wildlife resources were in great disaster and great threats. Carrying out practical and effective protection was imminent.This study prepared the data by means of data collection, spot investigation, data finishing and counting. Meanwhile, it determined 6 indexes including wild plants richness, wild animal richness, diversity of ecosystem types, extent of threatened species, protected species richness and extent of biological invasion to assess the diversity of wildlife resources based on the special status of study area. The results showed that there were 555 species of wild vascular plants,344 species of wild vertebrate,6 species of different ecosystem,289 species of threatened,50 species of state protection and 7 species of invasion. The consequence indicated that BI presented general, which was lower than national average level.According to the status of wildlife resources, the study analyzed problems in present protection and their causes. Also, it offered concrete protection plan:establishing a wildlife protection network in pan Dunhuang, which combined economic, society and culture. It contained special conservation strategy; building unified management organization in pan Dunhuang; comprehensive development and utilization of historical culture, geology and geomorphology remains; key protection of wildlife by building national desert wild germplasm resource pool and western aboriginal animals and plants garden, according to division method of three reserve group.It put forward division method of three reserve group as follows, based on the principles of adjacent position and same protected objects:1) the first, Dunhuang Xihu nature reserve, Dunhuang Yangguan nature reserve and Annanba Camelus ferus nature reserve, main function was building national desert wild germplasm resource pool;2) the second, Yanchiwan nature reserve, Big Sugan lake nature reserve and Small Sugan Lake nature reserve etc., main function was protecting the whole water source eco-system;3) the third, Anxi extremely drought desert nature reserve, Shule river middle and lower reaches nature reserve and Mazong mountain Capra ibex nature reserve, main function was building western aboriginal animals and plants garden.
