Component Analysis Of Metarhiziumanisopliae's Broth And Initial Abstraction Of Swainsonine

Objective: In orde to definite the main moity,initially, serial subcultivation and liquid state fermentation of Metarhiziumanisopliae was made.Obatining swainsonine from the seco- ndary metabolite in the Metarhiziumanisopliae's broth by some methods,such as centrifuging, separating,extracting and cleansing. To search for limpid abstaction approach,a new kind of t- reating tumor was expected.Method: (1) PDA was used for Metarhiziumanisopliae's passage and the strain was preserved under 4℃. To collet the broth after Metarhiziumanisopliae was cultured on Czape- k's solution.(2) Moity analysis of the broth did in accordance with plants-component analysis.(3) Pre-disposal treatment of the broth was made by high speed centrifugation,micro- bore-filter,resin-adsorbing.And then,to repeatedly extract by n-butanol, negative pressure chromatographic separating,reduced pressure sublimating,et al.on the condition of strong base (4) GC used to affirm the swainsonine in the sublimation.Result: The results showed that the Metarhiziumanisopliae grew well on the condition of 25~26℃,humidity 45~55%,pH6.5~7.0 and sufficient nutrient.Alkaloid(SW),amino acids,pro- tein,organic acid,tannin,Phenols,olein,aetherolea,Terpene,Steroid were the essential compone- nt of the broth.The characteristic color reaction of alkaloid was obviously.Characteristic color reaction and ultraviolet absorption curve of the sublimated component was the same as swainsonine control article.The results of GC showed that the appearance time and peak form of the sublimated component approached the swainsonine control article.Conclusion: There was swainsonine in the Metarhiziumanisopliae's broth.The method of this test saved detection time and dosage of reactant.Initial abstraction of swainsonine was completed.
