【Objective】The purpose of this experiment is to find a effective method in order to isolate swainsonine (SW) from Rhizoctonia lguminicola Culture Solution and to open up a new path for isolation awainsonine.【Methods】Rhizoctonia lguminicola were inoculated on PDA in a state time. Making it hold hearty vitality. Certain strains were cultured on Czapek's medium for culture solution. Swainsonine was detected with TLC in whole experiment. The last result was detected with gas chromatography.Deferent methods of isolation swainsonine from Rhizoctonia lguminicola: All methods include extraction with chloroform and n-butanol; Soxhlet extractor, elution and salvation with absolute alcohol; column chromatography with silica gel; and decompression sublimated. Finally, it appeared white powder. Following, five methods would be given.The first method: Culture solution (pH<4) that have been preserved was filtrated and filtrated again after adjusted pH>9 and precipitated. Filtrate was extracted with absolute ethanol (Soxhlet 8~12h) and extracted with n-butanol. N-butanol solutions have been extracted with acid solution (10% HCl). The acid solution was been evaporation to dryness after adjusted pH>9. Finally, it was eluted with absolute alcohol and decompression sublimated at 90℃after evaporation to dryness. It appeared white powder.The second method: Culture solution (pH<4) that have been preserved was filtrated and filtrated again after adjusted pH>9 and precipitated. Filtrate was extracted with n-butanol. N-butanol solutions have been extracted with acid solution (10% HCl). The acid solution was been evaporation to dryness after adjusted pH>9 and soaked with absolute alcohol in 24h. After these, it was been evaporation to dryness and decompression sublimated at 90℃. Finally, it appeared white powder.The third method: Culture solution (pH<4) that have been preserved was filtrated, and extracted with chloroform and n-butanol. Then, it was been evaporation to dryness after adjusted pH>9 with NaOH and extracted with absolute ethanol (Soxhlet 8~12h). Finally, it was isolated with column chromatography with silica gel (eluant is chloroform: carbinol: ammonia=70:26:2:2) and decompression sublimated at 90℃. It appeared white powder.The fourth method: It was been adjusted pH>9 again after the first method. Then it was been salvation with chloroform and decompression sublimated at 90℃. Finally, It appeared yellow extractum.The fifth method: Culture solution (pH<4) that have been preserved was filtrated, and extracted with chloroform and n-butanol. Then, it was been evaporation to dryness after adjusted pH>9 with NaOH and extracted with absolute ethanol (Soxhlet 8~12h). Finally, it was been salvation with chloroform and decompression sublimated at 90℃. It appeared yellow extractum.【Result】The first and third method were better than others. And their results were that it contained SW by TLC and gas chromatography.【Conclusion】It contained SW by TLC and gas chromatography. Although it contained a little of SW, it was to prove the method was useful. And these method need to further research and improve.
