Eucommia ulmoides Oliver,Do-zhong in Chinese, is a famous Chinses traditional medicine having good function in descending blood pressure. Pinoresinol diglucoside (PDG) has been known as the most important component contributing to descending blood pressure in Eucommia ulmoides Oliver.The traditional using method, peeling the bark or leaves, is greatly limited by the material because the tree is so special and has been listed in the second level of protected tree. Production of functional component with microorganism has many advantages in saving plant materials, having high efficiency and low cost. The thesis is aimed at getting the optimal conditions for production of PDG with a strain of Phoma sp., SP-16F, in liquid cultivation. Main contents and results are listed as follows.(1)By using Single-factor design, optimal conditions for cultivating seed was established as medium volume of 100ml in 250ml shake flask, initial pH of 8.1, rotation speed of 185r/min, temperature of 28℃, inoculum amount of 10 pieces, and cultivation time of 120 h.(2)Using Cebtral Composite Designed experiments and Response surface analysis, the fermentation medium for producing PDG was confirmed.The medium components were sucrose47.5g/L, NaNO33.0g/L, K2HPO42.83 g/L.(3)By using Plackett-Burman design and Box-Behnken design ,the fermentation conditions for producing PDG were established at temperature 28℃, inoculum amount 10%, rotation speed 193r/min, medium volume 100ml, initial pH8.4.(4) On the optimal conditions,we cultivated SP-16F in liquid medium for 216h, the highest yield of PDG were 30.51mg/L,and it is enhanced by twice to that before optimazition.(5)The established equations showed good fitness and prediction to the experimental values...
