In this paper, the Arabidopsis MSL gene was analyzed in the method of bioinformatics; the relationship between the regulation of the MSL gene family expression and the BR was studied using Realtime PCR in the Arabidopsis; and the over-expression vector of MSL gene was also built, so was the sub-cellular location vector and RNA interference vector. The result and conclusion are in below:The result of bioinformatics analysis showed that MSLl-3 have 5 TM domains, MSL4-10 are for 6. MSL1-3 contain putative N-terminal organelle localization sequences, and MSL1 is predicted to localize to mitochondria, MSL2 and MSL3 to plastids, according to the sub-cellular localization prediction program Predotar.The expression of MSL1 was not regulated by BR, and was not responded to the hypertonic stress. The expression of MSL9 was down-regulated by the endogenous BR, and so did by the hypertonic stress. The expression of MSL10 was up-regulated by BR, and the hypertonic stress did the opposite thing.The over-expression vector and sub-cellular localization vector of MSL1, MSL 10, MSL12 were built through the Gateway system. The RNA interference vector of MSL10, MSL12 was built using the RNAi. The interference vector of MSL1, MSL1/MSL11 was built using the Artificial miRNA.
