With the various types of dust and the toxic and harmful elements which in the working environment existing and exceeding, human health will be laid hidden danger, it had contributed to the terrible consequences of serious occupational diseases. Urea which was a kid of nitrogen fertilizer had high levels of nitrogen in the solid nitrogen fertilizer, but there were a large number of urea dust in the production process of the urea and packaging establishments in the air, soil, water and other places, it can cause human respiratory disease and kidney disease, and that irritates the eyes and the skin, and it is directly harmful to human health. Nickel was widely used in steel,alloy,oil industry, its various compounds were carcinogenic and were serious impact on the body's immune system,hematopoietic system,reproductive system and skin. Nickel and the concentration of urea in workplace air were very important indicators of human health, and determination of its content provide the vocational and non-professional populations with the advance detection target, and there had a certain reference value for the diagnosis of occupational diseases.So, accurate determination of urea in the air to the workplace and the trace nickel in human urine is very important. The high-performance liquid chromatography has great advantage on separation and determine of complex samples, it has a good effect on the selectivity and separation efficiency. But for the most of analysis of heavy metal, Graphite furnace atomic absorption spectrometry is the best analysis method because of it's high selectivity and high sensitivity. This topic research and establish the methods of determination urea in the air to the workplace and the nickel in human urine by graphite furnace atomic absorption spectrometry. The main research as follows:1. Determine urea in the air to the workplace use of high-performance liquid chromatography. Use C18 chromatographic column, discussion the opt of mobile phase, optimization of instrument conditions which included the flow rate of mobile phase, wavelength, column temperature. We also discuss the elution time of millipore in air sample collection, preclude the interference of other components and ammonia in the air. Under the optimum experimental conditions, the linear dynamic range of the calibration curve is 0-300μg/mL, the linear constant coefficient is 0.9996, the detection limit is 0.4μg/mL. It can meet determine urea in the air to the workplace.2. Establish a new method of rapid determination of nickel in the urine by atomic absorption spectroscopy, Focuses on the pre-treatment method of sample, text the impact of nickel in the urine with different acids and acidity, analyse the effect of different matrix modifiers and choose one matrix modifier, optimize it's use level, reduce the various kinds of interference of urine matrix to the nickel determine, optimize equipment and reaction conditions which include ashing temperature, atomization temperature. Under the selected experimental conditions, nickel has a linear relation within the scope of 0-5.00μg/L, the linear constant coefficient is 0.9998, the detection limit is 12μg/l, the recovery rate is 91.3%-105.3%. The method is simple and practical, stability,.It can provide a accurate and reliable analytical method of biological samples for laboratory tasting.
