The high performance capillary electrophoresis (CZE) and micellar elecyrokinetic capillary chromatogrphy (MECC) are applied to five medicine separation and analysis in this thesis. This investigation is very useful for expanding the application of CZE , MECC and developing analytic method for medicine. The thesis consist four chapters and main content as follows:l.To establish an accurate method for determination of genistein and 2,5-dihydroxybenzene sulfonate by HPCE.The CZE are applied to the determination of genistein in huaijiao pill according to experimental condition as follows: the quercetinum as internal standard substance, 20 mmol/L phosphate buffer (pH9.2), A =254 nm, and a content voltage of 28 kV. The experimental results show that the calibration graph is linear from 31.3 to 109.4μ g/mL(Y = 0.04149+0.01524C, r = 0.9994), and the average recoveries is 100.1% for genistein (n=3).The CZE are applied to the determination of 2,5-dihydroxybenzene sulfonate in calcium dobesilate capsules according to experimental condition as follows: the caffein as internal standard substance, 20 mmol/L phosphate+10 mmol/L borax buffer (pH8.2), r=214 nm, and a content voltage of 25 kV. The experimental results show that the calibration graph is linear from 37.9 to 75.7μ g/mL(Y = 0.13863+0.00493C, r = 0.9978), and the average recoveries of 2,5-dihydroxybenzene sulfonate is 100.4 % for 0.119mg/10mL 2,5-dihydroxybenzene sulfonate and 99.8% for 0.289mg/10mL 2,5-dihydroxybenzene sulfonate.2. The isomers of daidzein sulfonic sodium and chiral isomers of nateglinide areseparated by high performance capillary electrophoresis.The effect factors on the separation of two isomers of daidzein sulfonic sodium are investigated. The experimental results show that a good separation can be achieved in 50μrn×50cm untreated fused-silica capillary with 20.0 mmol/L KH2PO4+10.0 mmol/L borax buffer (pH 8.2) and 27kV applied voltage by the UV detector at 254nm. Because of the formation of boric acid ester between borax and daidzein sulfonic sodium, the isomers of daidzein sulfonic sodium are separated within 13 minutes .The effect factors on the separation of chiral isomers of nateglinide are investigated. The experimental results show that a good separation can be achieved within 16 minutes in 50μm×50cm untreated fused-silica capillary with 20.0mmol/L KH2PO4+10.0 mmol/L borax buffer (pH 8.45, containing 15% methanol and 30mmol/L DM-β-CD) and 25kV applied voltage, and detect wavelength of aUV detector is 210nm.3. The chiral isomer impurity of atorvastatin is separated and detemined by MECC. The effects of the concentration of SDS and HP-β-CD on separation are investigated, and the method of quantitative analysis is established. The experimental results indicate that the impurity content steady within the sample concentration range from 0.4g/L to 0.2g/L for atorvastatin.The author has finished four paper during the graduate research.
