Yd was screened in nature, which can produce a fibrinolytic enzyme. The fibrinolytic enzyme will be developed as a thrombotic agent. A series of treatment has been done to the initial strain Y32, to bread a new mutant梇c1. It's condition of characterization and fermentation was investigated.Y32 was identified as Rhizopus sp. by its morphology. The fibrinolytic enzyme activity of Yc1 reached 53.845UK/ml, increasing 63.8 fold.By the method of orthogonal test, the optimal main medium was achieved, which contained 5.0% wheat bran, 3.0 % soybean meal, 0.03% ZnCl2.0.15% MgS04.7H20, 0.05% Na2HP04.12H20, 0.6 %K2HP04.3H20, pH 7.0. The Yc1 was inoculated into 40ml medium in 250ml triangle flask shaking in 30℃ at 160rpm for 60h. The fibrinolytic enzyme activity of Yc1 reached 163.059UK/ml increased 202.83 fold.A fibrinolytic enzyme was purified from the fermented broth of Yd by ammonium sulfate fractional precipitation.
