Malachite green and gentian violet are triarylemethane dyes. They are highly effective against protazal and fungal infections. Malachite green and gentian violet are potentially.carcinogenic and have already been prohibited in many countries. Malachite green and gentian violet can be transformed into leucomalachite green and leucogentian violet respectively. The simultaneous determination of these four substances by HPLC was studied in this paper. The pretreatment of samples and the terms of HPLC were mainly studied. For this method, pretreatment of the sample is the key step. After comparison of various cleaning and enrichment methods, PRS SPE with a post column filled with Pb02 was applied because this approach was good for the good effect and good recovery. Recoveries from lOng/g fortified fish tissue were 77 + 1%, 80?%, 89 + 3% , 90 + 1% for MG, LMG, LGV and GV respectively (mean S. D. , n=4).It was noted that previous residue determination often only focused on the determination of the original medicine. Both the original medicine and its major metabolite were determined with a derivative post-column in this experiment. Consequently, it could be better used to monitor the illegal use of the medicine and guarantee the aquatic products safety. After modifying the HPLC terms, all of these four substances can be simultaneously determined without interference. This multi residuedetermination method can also be beneficial to the job of the inspection agents.Based on our investigation of the aquatic products residue, no malachite green,gentian violet and their metabolites residue were found in most samples. However, one sea perch sample was found to have a high leucomalachite green residue.The metabolism of malachite green was also studied. Result showed that malachite green could not only be transformed into leucomalachite green but also be transformed into other metabolites. Malachite green decreased quickly while leucomalachite green persisted for a long time. Leucomalachite green should be targeted in residue-monitoring programs.In general, we studied: the simultaneous method of determination of malachite green N gentian violet and their metabolites; the persistence of malachite green and leucomalachite .green in fish tissue; the malachite green and gentian violet residue in Qingdao local markets. The technical determination method and the study on the persistence which was provided in this paper can contribute to the residue-monitoring programs.
