[Background]The development at full speed of modern science and technology and social economy, make people's living standard improve constantly, life quality improve greatly, and the human health has already become one of the themes studied in our times. People are more and more urgent to green healthy medicines and green safe food, and to develop and utilize the natural plant to bring benefit for mankind, which is the present developing trend. Our country is a big agricultural country, animal husbandry is growing vigorously, and it is important problem to control beasts and poultry diseases. At present the majority of medicine and feed additives used in the animal husbandry is antibiotic. The antibiotic has important functions in promoting animals to grow and reducing the incidence of suffering from diarrhoea for many years, but in recent years, with improvement of living standards of people and growing with people's food security and environmental protection of consciousness, people realized that using antibiotic makes the drug residue more and more seriously and had already caused the environmental pollution and threatened people's health. Some developed countries and regions in the world have already taken measures to forbid and restrict them gradually, and many countries and regions are devoting to develop and popularize the environment-friendly green products now. So, to develop and utilize abundant plant resources rationally in our country, and substitute the antibiotic with materials extracted from plant, it is the unavoidable trend and one of the important subjects that the contemporary scientific workers face.[Objects]Study the extractive technology of Mosla Chinensis Maxim volatile oil (MCMVO), and identify its chemical compositions; Carry on security appraisal to MCMVO, and study the antibiotic and antivirus effects and immunoregulation; Study inclusion technology for inclusion compound of MCMVO.[Methods]The MCMVO was extracted by supercritical CO2 and organic solvent method, and its chemical compositions were analyzed by gaseous chromatogram-mass spectrum (GC-MS). With mice as materials, acute toxicity test and subchronic toxicity test were observed, and the lethal dose 5O(LD50) of mice were analyzed; The antibacterial effect of MCMVO was measured by the cup-plate method, its minimal inhibitory concentration (MIC) and minimal bactericidal concentration (MBC) were measured by double dilution tests, and its antiviral effect was measured by conventional methods. The carbon clearance assays, delayed type hypersensitive andstomach wriggling tests were selected to observe the effects of MCMVO on the immune functions. (3-cyclodextrin inclusion compound of MCMVO was prepared by saturated aqueous solution method, and the orthogonal test was used to study preparation technology in P-cyclodextrin inclusion compound of volatile oil.[Results]1. The MCMVO was extracted by supercritical CO2, and its extraction rate was 3.4%. The best extraction technology of organic solvent method: To extract volatile oil dividing two steps, total time is 210 minutes, the total proportions of input of the solvent and raw materials are 8:1 (g:ml), and temperature is 65"C, the extraction rate is 1.85%.2. GC-MS is used to analyze volatile oil, the results suggested the materials in Hunan contain phenols, alkenes, esters, fatty acids, alkanes, alcohols etc. The main composition is thymol, carvacrol, P-cycmene, humulene etc. The thymol has the highest content accounts for 56.25%, and secondly the carvacrol is 19.21%.3. The result of acute toxicity test shows : LD50 of MCMVO to mice is 1.303±0.116 ml/ kg'bw, 95% interval range is 1.419-1.197ml/ kg'bw.4. The results of subchronic toxicity test show: The MCMVO has not obviously made the mice grow, and no obvious influence on the hemoglobin(HB), red blood cell (RBC), white blood cell(WBC) and its counter; MCMVO has the effect to reduce the blood total cholesterol (TCHO) and blood urea nitrogen (BUN), and has no influence to the other biochemical index of blood; MCMVO has no obvious influence to organs weight and organs index of mice.5. In the experiment of antibacterial activity against Escherichia coli, Pasteurella, staphylococcus aureus and streptococcus, the MCMVO has stronger antibacterial activity than that of Berberine sulpuate (1%), and is no evident difference comparing with Lincomycin hydrochloride (5%) and volatile oil of Origanum Vulgare L.(6%). The MIC and MBC of MCMVO to E.coli, Pasteurella multocida, staphylococcus aureus and streptococcus are 0.625 mg/ml, 0.625 mg/ml, 0.312 mg/ml, 0.312 mg/ml and 2.50 mg/ml, 5.00mg/ml, 1.25 mg/ml, 2.50 mg/ml.6. 0.79 g/L and 0.39 g/L dose group of MCMVO have the function of anti-NDV notably , and is stronger than 1.0 g/L of virazole.7. The MCMVO can improve carbon clearance index and phagocytic index of mice, and has obviously regulating effect on non-specific immune function; The MCMVO make DTH action of skin strengthen which is caused by DNFB, and improve the specific immune function of normal mice; The MCMVO can increase weight of chest gland and spleen of normal mice.8.
