Zizyphus jujube date is fruit of Zizyphus jujube Mill(Rhamnaceae), also named Chinese jujube date, red dates. It is mainly planted in China, has high nutritious value and excellent pharmaceutical function and is a necessary component in traditional Chinese medicine. The amino acid, Vitamin,mineral,cAMP,cGMP and some other nutritional component have been investigated deeply. However the research work on polysaccharide from jujube date is very limited, especially in our country, such research work is just in initial phase. In this thesis, the sugar component of Chinese jujube date have been studied, the extraction, isolation, purification, gradation, purity identification, physical and chemical behavior, structure research of jujube date polysaccharide(shorted in JDP) have been researched in detail.Water-soluble polysaccharide from Chinese jujube date is extracted and several factors affecting extraction are discussed. The condition suitable for extracting Water-soluble substance of jujube date is : temperature 80°C; three times, each for 7h,2h,2h respectively; ratio of water and jujube date is 3:1~10:1.During extraction, jujube date should be properly crashed and agitated. Dry weight of extraction liquid accounts for 67.9% of jujube date flesh(dry wt.). Ultrasonic wave can accelerate extraction process; In jujube date water-soluble extract, polysaccharide accounts for 1.1698%.During de-protein process, Sevage method, several kinds of proteinase methods have been compared. During de-color process, active carbon, iron exchange resin have been used. Two proteinase method is more suitable for de-protein of crude JDP, and iron exchange resin is more suitable for de-color. Dialysis by running water and distilled water can isolate most of low molecular substance and ion.JDP was graded by DEAE-cellulose method. The DEAE-cellulose column was eluted with water, followed by gradient elution with potassium acetate solution. The elute with water was neutral polysaccharide(named JDP-A). Purity was identified by gel chromatography.JDP-N was white powder. JDP-A is light yellow powder. Structure of JDP was studied by Infra-Red spectrum, Ultra-Violet spectrophotometry, component sugars of two kinds of JDP are all a-furan sugar.
