Polycyclic Aromatic Hydrocarbons (PAHs) is a kind toxic organic contamination distributing in environment abroad and difficult to be degraded by biology. In environment it is easy entrance organism and sediment, and though food chain enter into body of human which can make serious latency harm to human health and ecology conditions. Bioremediation is a import method treating contaminated by PAHs. The bacteria of An2 has high capability for degrading PAHs and Pseudomonas aeruginosa has preferably capability for producing biosurfactant. After An2 was mutagened with low energy N~+ implantation and Pseudomonas aeruginosa was markered with EGFP, carry through cell electrofusion, the Genetic Recombination Microorganisms (GRMs) which has more capability for degrading PAHs was gained. The whole work includes:1. Up primer P1 and down primer P2 were designed according as enhance green fluorescent protein (EGFP) gene .With the study of Pseudomonas aeruginosa markered with EGFP, constructed the system of Pseudomonas aeruginosa marker, recombined strain of P. aeruginosa/EGFP was gained .The capalitity of EGFP marker was stabilization.The strain detertined by genomic DNA PCR and spot blotting, the transformed EGFP gene lied in chromosome of Pseudomonas aeruginosa2. Though the bacteria of An2 mutagened with low energy N~+ implantation, the mutant strain of An815 was gained. The capability of An815 for enduring and degrading anthracene improve greatly than An2.3. In the work, for discuss the affestment on preparation and regeneration of Pseudomonas aeruginosa and An815 protoplast, confirm the best condition of preparation and regeneration of Pseudomonas aeruginosa and An815 protoplast,through made cross-test experiments on preparation and regeneration of Pseudomonas aeruginosa and An815 protoplast.4. Though protoplasts of and An815 and Pseudomonas aeruginosa/EGFP electrofusion, the fusant R2 was gained, which had the capability not only for Degrading Polycyclic Aromatic Hydrocarbons of An815 ,but also for producing biosurfactant of Pseudomonas aeruginosa. The capability for Degrading Polycyclic Aromatic Hydrocarbons was improved greatly.5. Though examination of shaking bottle aerobic culture, with fusant R2 improve general biology treatment, the bioaugmentation degrading coke-plant waste water was studied. The COD removing rate of coke-plant waste water by amend sludge added fusant R2 up to 66.4%, it increase 16.2% than comparison. The COD removing rate of activated sludge was improved by added fusant R2. Experiment shows that fusant R2 is potential in degrading cok-plant waste water.
