| Lipase from Candida sp. is one of important microorganism enzymes in industrial application. In the present work, the fermentation production of lipase with Candida sp.99-125 was studied. We began with establishing the method to determine biomass and concentration of fat metabolic intermediate. After that we optimized the medium by trying to replace soybean meal with soybean meal hydrolysate or bean flour. The lipase activity using soybean meal hydrolysate as nitrogen source reached 8500IU/ml and bean flour was also believed to be the perfect substitution of soybean meal.Then the oil adding strategy was well studied. Shake flask culture was taken to show how Candida sp. is consuming oil and three different oil adding strategies was compared to each other. After that the adding strategy that adding rate be direct proportion to the lipase activity which assured the stable concentration of oleic acid in fermentation liquor and thus assured the stable high lipase activity was chosen.At last we competed some basic works of the relations between the component and function of lipase. Method to determine the esterification activity of lipase was established and the activity of lipase fermented using different carbon source was compared. |
