Sucrose-containing polymer means that leading sucrose to the polymer molecular chain group to form macromolecular polymers.Due to the integrated sucrose molecule configuration,sucrose-containing polymer has the hydrophilicity,biological activity,biodegradability.It has the applied foreground in the hydrophile material,drug controlled release material field.There are three methods to prepare the sucrose-containing polymer: enzyme method,chemical method,pre-irradiation method.The chemoenzymatic method combines the high regioselectivity of enzyme method and high polymerization efficiency of chemical method,it is the ideal method to prepare sucrose-containing polymer.In this paper,proteinase N catalyzes synthesis of sucrose methacrylate,the monomer of sucrose-containing polymer,in the pyridine and DMF.In pyridine, use the different pH carbonate-sodium hydroxide to prepare the pH memory proteinase N.Trifluoroethyl methacrylate is used as acyl donor,sucrose as lyoprotectant.Study the effects of reaction dosage ratio,reaction time, temperature,system H2O content,pH of proteinase N to the concentration of sucrose methacrylate.Design the orthogonal experiments,the optimum conditions were found using pH 8 and 1.5%H2O and 35℃and 5days.The highest concentration of sucrose methacrylate 9.85%was obtained in this condition.In DMF,use the different pH phosphate buffer to prepare the pH memory proteinase N.Trifluoroethyl methacrylate is used as acyl donor,sucrose as lyoprotectant.Study the effects of reaction dosage ratio,reaction time,temperature,system H2O content,pH of proteinase N to the concentration of sucrose methacrylate.Design the orthogonal experiments,the optimum conditions were found using pH 12 and 7%H2O and 35℃and 3days.The highest concentration of sucrose methacrylate 22.15%was obtained in this condition.Identify the structure of sucrose methacrylate by IR,MS,13C-NMR,ensure that:ester exchange reaction happened in the 1'C of sucrose.K2S2O8/H2O2 and Ce(NH4)2(NO3)6 polymerize the sucrose methacrylate monomer respectively to get the sucrose-containing polymer in the normal temperature,study the effect of dosage,reaction time to the molecular weight of polymers and the translation rate of scrose methacrylate monomer.Molecular weight of sucrose-containing polymer is from 5,000 to 600,000.The application of sucrose-containing polymer on drug controlled release, the drug could be slowly released in 12h steadily.It has better release effect in man-made intestinal juice than in man-made succus gastric.
