Endophytes of Melia azedarach L. are of great importance to agricultural and medicinal research for their generation of diverse secondary metabolite products not only antibacterial and insecticidal activity but also environmental compatibility. Mixed fermentation attracted more attention for having more advantage over pure breed fermentation. Based on this and for the purpose of finding novel bioactive compounds from Melia azedarach L., mixed fermentation of endophytes in Melia azedarach L. was carried out,the study on Chemical Constituents and bioactivity of mycelium from mixed fermentation was also presented in this thesis。The mixed fermentation of endophytes LN-14and LN-15 in Melia azedarach L. was employed to produce mycelium,of which the ethyl acetate extract was subjected on silica column chromatography to generate 8 fractions(Fr.1~ Fr.8), and the detection indicated that margosine was concentrated in Fr.4 and Fr.5.The Anti-bacterial (E. coli, P. aeruginosa, S. aureus, B. subtilis, B. cereus) , fungicidal (G. cingulata, B. cinerea, A. solani, F. oxysporium, V. alboatrum, F. solani, C. lunata, F. graminearum) and insecticidal activities ( 3rd instars larvae of Mythimna separata Walker) of ethyl acetate extract and Fr.1~ Fr.8. it has been observed that Fr.4 and Fr.5 exhibited good activity against the tested bacteria and fungi, the EC50 values of Fr.4 and Fr.5 against the mentioned 8 fungi were 0.8592,0.5316,0.5873,1.5947,1.2348,3.3806,2.0145,9.3230 mg.mL-1 ,and 1.0447,0.5015,0.5281,1.5011,1.3516,3.9959,1.1664,2.9855 mg.mL-1, respectively.Five compounds were isolated from ethyl acetate extract, one of which was indentified as (2R)-2-hydroxy-N-((2S,3R,4E,8E)-3-hydroxy-9-methyl-1-((2R,3S,4R,5S)-3,4,5-trihydroxy -6-(hydroxymethyl)tetrahydro-2H-pyran-2-yloxy)octadeca-4,8-dien-2-yl)hexadecanamide, that is Cerebroside B, on the basis of 1HNMR, 13CNMR, and ESI-MS analysis and through the comparison with literature. That is the first report on the structural elucidation of this compound from endophytes in Melia azedarach L.
