Capillary electrophoresis(CE) technology is a kind of high efficiency, high sensitivity, rapid biological macromolecular separation and analysis tools. It has more great advantage for the separation of protein analysis, however, the protein adsorption on the inner wall of the quartz capillaryin the process of analysis of makes separation analysis not accurate.The most main and commonly used method to solve this problem is to fuse quartz tube wall coating processing today.So far, this kind of coating method is mainly divided into physical adsorption and chemical bonds coating and each have advantages and disadvantages.How could combine the advantages of these two kinds of coating and avoid the shortcomingsperfectly to prepare high performance, high quality capillary coating to achieve betterprotein separation analysis result is a key point that researchers in this field need to breakthrough.This paper use the layers by layers(LBL) self-assembly technology and security environmental protection water soluble photosensitive polymerdiazoresineinstead of the strong toxicity, polluting silicon alkylation reagents. This molecule has a unique cross-linking reactionof optical and thermal.And then realize the capillary wall coating and immobilized self-assembly covalent bonds. The preparation of the coating is used to analysis the separation of proteins and it has achieved good results.This thesis mainly works as follows:1. We use the photosensitive polymer diazoresin(DR) as the coupling agent, through self-assembly and covalent connectivity CD-Dendrimer for capillary coatings.Diazoresin and CD-Dendrimer fixed by ionic bonding on the inner wall of the capillaryfirst and then ionic bonds turn into covalent bondsthrough UV light exposure, thereby it could fix on the inner wall of the capillarystability.This kind of coating to separate several mixed protein, it shows a good separation effect.2.Prepare the diazoresin and Graphene Oxide(GO) capillary coating on the inner wall of the capillarybased on the layers of self-assembly(LBL) technique.Two kinds of molecular coat alternately repeatedly and the weak interactions between the graphene oxide ionic bond turn into stronger covalent bondsunder the irradiation of ultraviolet lightfinally.Thereby prepared multilayer structure controllable covalent bond and coating used to separate several alkaline protein, the separation effect is ideal, reproducibility and stability is also very good.3. Select the hydrophilic substances β-cyclodextrin(β-CD) as the separation of protein materials and diazowith resin to form capillary surface composite membrane by ionic bonds tohave the effect of coated capillary.Then take it under uv irradiation, the ionic bond turn into stable covalent bondthrough the unique optical response. The capillary coating is more stable and not easy to fall off.The inhibitionof protein adsorption is obvious. The prepared of covalent bond DR/β-CD capillary coating for basic protein separation, the effect is very good.In this paper the research and preparation of the capillary coating layer is a combination of the advantages of physical adsorption and chemical coating two methods by light cross-linking.They area kind of controllable and stable covalent bond formation structure and coating.The experimental results of this paper is proved to be that the coating preparation technology is a kind of simple preparation, high efficiency, good reproducibility and green environmental protectioncapillary method. |
