Changium smyrnioides Wolff belong to Umbelliferae family. The root of Changium smyrnioides Wolff is a kind of precious Chinese crude drug. It has the functions of relieving cough, moistening lung, regulating immune, promoting digestion and absorption etc. In order to make further use of the Chinese herbal resources, the extraction, purification and structure characterization of the chemical constituents from Changium smyrnioides Wolff were studied. The antioxidation and hypoglycemic activity of the extracts were also studied. The research contents are as follows.Changium smyrnioides Wolff 10kg was extracted with heating reflux using 95% ethanol as the solvent. The extracts which were condensed under reduced pressuret were extracted with petroleum ether, ethyl acetate and n-butanol. Then 103 g petroleum ether extract,88 g ethyl acetate extract,330 g n-butanol extrcat and 1000 g water extract were obtained. On the basis of preliminary study, the antioxidation and hypoglycemic activity of diferent extracts from Changium smyrnioides Wolff were studied. Scavenging activity against DPPH and inhibition on a-glucosidase were detected.The results showed that the ethyl acetate extract has favourable antioxidation and hypoglycemic activityThe chemical constituents from Changium smyrnioides Wolff was studed. Column chromatography was used to separate the petroleum ether extract. The solvent system petroleum ether-ethyl acetate were used to gradient elution. The fraction was detected by TLC. Two compounds were obtained. They were identified as lignoceric acid and daucosterol. HSCCC was used to separate the ethyl acetate extract and n-butanol extract. The solvent system chloroform-methanol-water (4:3:2,v/v/v) and ethyl acetate n-butanol-water (4:1:5, v/v/v) were chosed. Three compounds were obtained from every polar fraction. The purification was detected by HPLC. The compounds which from the ethyl acetate extract were identified as 8-methoxy-5-hydroxy psoralen, 1,4-succinic acid and p-methoxycinnamate glucoside. HSCCC is considered as a suitable alternative for the separation of natural product due to its unique advantages, especially some compounds difficult to isolate by the other chromatography. HSCCC is an effective complementary way to other conventional methods for isolation of chemical constituents from traditional Chinese herbs.
