Euphorbia sieboldiana Morr. et Decne. belongs to the Euphorbia L.,Euphorbiaceae. It widely distributed in many provinces of China. It has a long historyof medicinal use in Tibetan medicine, and has many kinds of pharmacologicalfunctions. Its water extract, ethanol extract and acetone extract have excelledinsecticidal activity against several insects such as Aphis sp. in Lycium barbarumLinn., Pieris rapae, and so on. This paper provided valuable experimental data forfurther development of this plant.This thesis selected the roots of Euphorbia sieboldiana produced in Qinghai asthe investigative object. By using normal chromatographing methods and high speedcounter current chromatography,11compounds were obtained from the petroleumether fraction and dichloromethane fraction of the methanol extract.High speed countercurrent chromatography was successfully applied to differentfractions of the methanol extract in different two-phase solvent systems composed ofpetroleum ether (P)-acetic ether (E)-methyl alcohol (M)-water (W). P:E:M:W=12:1:8:1, petroleum ether fraction was divided up6fractions (Fr1, Fr2, Fr3, Fr4, Fr5and Fr6). By using HSCCC, these fractions were isolated and sublimated again.P:E:M:W=5:5:5:3, compound1and2were isolated from Fr1. P:E:M:W=3:1:2.6:0.4, compound3was isolated from Fr3. P:E:M:W=4:1:2.5:0.5, compound4and5were isolated from Fr4. P:E:M:W=6:1:3.8:0.4, compound6,7,8and9wereisolated from Fr5, compound10were isolated from Fr6. P:E:M:W=5:5:6:3,compound11was isolated from dichloromethane fraction.On the basis of their physical constants,1H-NMR,13C-NMR and MSspectroscopic data, modern analytic techniques and necessary chemical reactions, thestructures of8compounds were elucidated, which covered diterpenoids,triperpenoids, flavones, and so on. Among them, three compounds (5,9and11) wereisolated first from this plant and six compounds (2,3,4,7,8and10) were known.11compounds were:1-octacosanol (2), β-sitosterol (3), ingenol (4), Jolkinolide B (5),atis-16-en-13(s)-hydroxy-3,14-dione (7), piperitol (9), β-amyrin acetate (10) and quercetin (11). Compounds4,5and7are diterpenoids. Compound9is volatile oil.Compound10is triperpenoid. Compound11is flavone.
