Background:As the material life rich increasingly, people pay more attention to the external image and internal mental health than ever before. Studies have shown that problems such as alopecia and premature canities can affect the mental health and the quality of patients’life. People’s hair has not only have the biology and physiology function such as defense and heat preservation, at the same time also has the effect of psychology and sociology. The incidence of hair problems rised year by year and the occurrence of hair problems have the trend of rejuvenation. Hair health problems cannot be ignored. At present most of the clinical medicine is chemical synthetic, cannot cure alopecia completely and along with side effects. There is no effective drug for treatment. "Yizhihao", Xinjiang Uygur traditional Chinese herbal preparation, has a definite therapeutic effect on alopecia. "Yizhihao" is consist of the seeds of Nigella glandulifera. Radix Polygoni Multiflori, Cacumen platycladi and so on. This paper got the financial support from Xinjiang Uygur science and technology project (No.2013911011) and Innovation Team Project Foundation of Beijing University of Chinese Medicine named’Lead Compounds Discovering and Developing Innovation Team Project Foundation’(No. 2011-CXTD-15).Research purposes:To study the effects of Xinjiang Uygur traditional Chinese herbal preparation "Yizhihao" on mice skin histology and hair growth in mice, and to investigate its possible mechanism. Formulating its quality standards is in order to ensure the quality of "Yizhihao". This issue aimed to provide scientific experimental evidence and data support for its clinical application, explore the possible mechanism of hair growth promotion, promote the development of its industrialization and provide a natural, safe and effective drugs and products for patients.Research methods:Kun Ming mice were chosen to be hair removal model by using Na2S as experimental animals. The hair growth condition was observed, as well as the hair length and weight of each group. In order to investigate the effect of "Yizhihao" on the tyrosinase activity in vitro by measuring the oxidation rate of L-DOPA. The main active fraction and components of "Yizhihao" were isolated and identified by HPLC-EI-MS. MTT assay was used to investigate the proliferation effects on human umbilical vein vascular endothelial cells (HUVECs) and PC12 of extract from "Yizhihao" and TSG. Taking the ratio of GSH and GSSG as the research object is to study its effect on the intracellular redox balance system in PC 12. According to ’China Pharmacopoeia’ 2010 Edition, TLC and HPLC methods were used in the determination of TSG in order to ensure the quality of "Yizhihao". On the basis of HPLC-ESI-MS analysis seven compounds have been dentified as gallic acid, emodin, 2,3,5,4’-tetrahydroxystilbene-2-O-glucoside, emodin-8-O-β-D-glucoside and so on.Results:Compared with the normal control group, the hair length, weight and the number of hair follicles showed great significant difference (P<0.01,P<0.001). Compared with the positive control group, the hair weight was higher and the number of hair follicles was more in the test group. None of the test groups could activate the tyrosinase activity effectively. Conversely, The water extract, acid extract of "Yizhihao" and TSG can not signifienatly deereased the activity of Mushroom Tyrosinase. TSG inhibited the tyrosinase activity in a concentration dependent manner at a concentration of 12.5~50 μg/mL. TSG exhibited a significant tyrosinase inhibitory activity (reached 32.87±8.81%) at the concentration of 50μg/mL. "Yizhihao" could promote the growth and proliferation of HUVECs significantly in the range of 7.3125~125 μg/mL. The maximum proliferation rates of "Yizhihao", acid extraction and TSG were 28.1%,45.7% and 80.4%. Gallic acid inhibited HUVECs growth in a concentration dependent manner at a concentration of 7.3125~125 μg/mL. Water extract and acidic site with increasing concentration of administration, the cell proliferation rate decreased in 125~1000 μg/mL, showing at 500μg/mL from inhibition. Combined with HPLC profiles, we can know tha acid site and TSG is the main active site and the active ingredient. "Yizhihao" and TSG could promote the growth and proliferation of PC12 significantly in the range of 7.3125~125 μg/mL. "Yizhihao" and TSG can protect PC 12 cells from oxidative stress induced by CoCl2.Conclusions:Xinjiang Uygur traditional Chinese herbal formulation "Yizhihao" can significantly promote hair growth in mice and its possible mechanism is to promote the growth of hair follicles, HUVECs and PC 12. The mechanism may be to improve microcirculation, promote the growth of hair follicle, protect nerve tissues and adjust intracellular redox equilibrium. Study on quality standards benefited for writing Yizhihao’s quality standard draft.
