| Objective:Observe aquaporins’expressions on the digestive system and brain tiss ue in chronic stress Liver Qistagnation and Liver-stagnation and Spleen-deficien cy model rats,and the expressions of motlin,gastrin in blood,discuss the the pat hogenesis of Liver Qistagnation and Liver-stagnation and Spleen-deficiency.Method:50behavior score-closed adult SD male rats (180g approximately) were divided into5groups randomly:(1) normal control group;(2)Liver Qistagnation group (replicated by chronic unpredictable mild stress model.);(3) Liver-stagnation an d Spleen-deficiency group (chronic stress+excess fatigue and improper diet gro up);(4) Liver Qistagnation+Chinese medicine intervention group (chronic stress plus Chinese medicine intervention)(5) Liver-stagnation and Spleen-deficiency+Chinese medicine intervention group (chronic stress, excessive fatigue and inte mperance diet, plus Chinese medicine intervention); Each group contains10rat s. from the7th day of making model,two Chinese medicine group s were intragastric administration Xiao Yao Wan once everyday; model group s are served with distilled water; normal groups are without any stimulation, n atural breed. Observation rat ethology score, by taking the brain organization, g astrointestinal tissues to detect the contents of corresponding indexes and blood hormone levels were compared, the whole process general situation of rats.Result:1.There’s no obviously change of AQP4on the rats hippocampus in each group (p>0.05).2. The expressions of AQP3on the small intestinal mucosa of liver-stagnation a nd spleen-dificiency were down-regulation,after TCM intervention expressions u p-regulation, compared with normal control group (P<0.05),expressions of Live r-stagnation and Spleen-deficiency group were down-regulation than Liver Qista gnation group (P<0.05);on the colonic mucosa,the expressions in each group were down-regulation than normal control group (P<0.05);the expressions of L iver-stagnation and Spleen-deficiency group and Chinese medcine control group1were down-regulation than Liver QiStagnation group (P<0.05);compared wit h Liver-stagnation and Spleen-deficiency group,the expressions of chinese medci ne control group2were up-regulation (P<0.05).3.The expressions of AQP8on the gastrointestinal mucosa in Liver-stagnation a nd Spleen-deficiency group were down-regulation than normal group and Liver Qistagnation group (P<0.05),after chinese medcine intervention expressions were up-regulation, compared with normal group and Liver-stagnation and Spleen-de ficiency(P<0.05); AQP8’s expression on the gastrointestinal mucosa in Liver Qi stagnation group and chinese medcine group1were up-regulation than normal group(P<0.05), expressions of AQP8on the colonic mucosa were down-regula tion than normal group (P<0.05).4.Liver Qi stagnation and spleen deficiency group, group MOT, GAS were lower tha n the normal group (P<0.05), two GAS contents in Chinese medicine group compare d with the model group increased significantly (P<0.05), compared with the normal g roup had no difference.Conclusion:1.There’s no obviously change of AQP4on the rats hippocampus in each grou p, shows that the occurrence and development of depression may not affect the wat er metabolism of the brain.2.AQP3in the group of stagnation of liver Qi with deficiency of the spleen in the inte stinal mucosa of expression in varying degrees down and reduction of stagnation of li ver Qi syndrome is not obvious, after Xiaoyao Pill intervention can improve the expre ssion, suggest depression of water metabolism in the development, major relevance an d liver stagnation and spleen deficiency syndrome, and suggests water wet disorder oc curs mainly in the small intestine and colon. 3. AQP8in stagnation of liver Qi with deficiency of the spleen group in gastrointestin al mucosa of expression were down regulated and Xiaoyao Pill stem prognostic up-re gulated the expression, suggesting that AQP8expression downregulation may for dep ression of liver stagnation and spleen deficiency syndrome, spleen governing transpor tation of hypofunction causes one of the mechanism of water metabolism disorders.4. Models of gastrointestinal hormone in rat serum MOT, downregulation of GAS exp ression, and the traditional Chinese medicine intervention upregulate the expression, s uggesting that depression of liver qi stagnation, Liver stagnation and Spleen deficienc y existed in functional gastrointestinal disorders. |
