The research took Coreopsis tinctoria flowers as raw materials,,study the best extraction technology of flavonoids in Coreopsis tinctoria, Separation and purification of Flavonoids from Coreopsis tinctoria at the optimal conditions by macroporous resin chrysanthemum, Coreopsis tinctoria extract liquid by freeze drying, the products obtained for further separation and purification of flavonoids by high speed countercurrent liquid chromatograph (HSCCC), obtained three high purity monomer compounds, and the structures were identified on the three compounds. To provide theoretical basis and technical support for further development and utilization of chrysanthemum.1Study on The Extracting Technology of Flavonoids from Coreopsis tinctoria flowersThe research took Coreopsis tinctoria flowers as raw materials, flavonoids were extracted from Coreopsis tinctoria flowers by using ethanol refluxing extraction. In order to determine the total flavonoids in extract, Rutin was used as a standard,510nm was chosen for detecting wavelength, and NaNO2-Al(NO3)3-NaOH colorimetric method was used. The results showed that the using testing method was accurate. Single factor test and orthogonal experiment results showed that the effect of extracting total flavonoids from Coreopsis tinctoria flowers size:Concentration of ethanol, extraction temperature, the ratio of raw materials to solvent, extraction time. The best extraction process conditions for Coreopsis tinctoria flowers were:the solvent60%ethanol, temperature90℃, the ratio of solid to liquid1:40, and the extraction time60minutes. The extraction result showed that the yield of total flavonoids can reach as high as11.23%.2Study on Technology of Separation and Purification for Flavones from Coreopsis Tinctoria Flowers By Macroporous Absorption ResinBased on the Coreopsis Tinctoria Flowers as raw materials, the purification process conditions of polyphenols from Coreopsis Tinctoria Flowers by macroporous absorption resin were studied. In order to set up the technology for adsorbing and separating Flavones from Coreopsis Tinctoria Flowers with macroporous adsorption resin,6types of resin were investigated. The results showed that NKA-resin was the best for adsorbing and separating flavones in the following technological conditions:the concentration of sample of extract, 6mg/ml; pH3.0-4.2; the flow rate of fluid,2ml/min; the eluting regent,60%ethanol,6folds the volume of the resin; elution rate,3ml/min, desorbing rate was95.5%, the maximum adsorbing capacity of NKA-2resin for Flavones was205.31mg/g, the content of product was82.83%.3Isolation and Preparation of flavonids from Coreopsis Tinctoria Flowers by High Speed Counter-Current ChromatographyA method for isolation and preparation of anthocyanins from Coreopsis Tinctoria Flowers by high speed counter-current chromatography (HSCCC) was established with using the biphasic solvent system water-n-butanol-ethyl acetate-TFA (5:1:3:0.0017, v/v).,upper phase acting as stationary phase, layer phase as mobile phase,850rpm,25℃, flow rate:2ml/min.
