The flavonoids of Coreopsis tinctoria was used as the research object in this experiment. The extraction, isolation and purification of flavonoids was researched, and the main contents were as following:1.The ultrasonic wave and ethanol extraction was used to extract the flavonoids from the Coreopsis tinctoria, and the experimental factors were concentration of ethanol, solid-liquid ratio and ultrasonic time. The orthogonal was conducted based on the single factor experiment, and the optimal extraction conditions of flavonoids from Coreopsis tinctoria by ultrasonic was that:the concentration of ethanol was 80%, the solid-liquid ratio was1:60, and the ultrasonic time was 1.5h. The extraction rate of total flavonoids can achieve 17.19 with three times in this condition.2.Two macroporous of AB-8 and D101 were selected in the experiment. The static adsorption and desorption performance of the two macroporous, the adsorption and desorption performance with different temperature, the static adsorption and desorption dynamics were researched in the experiment. The results showed that: the adsorption and Desorption effect of the two Macroporous was well, and the adsorption rate and desorption rate were more than 80%; the adsorption and desorption could achieve good level in 30℃, and the Process can complete mostly in 6~7h. The two macroporous of AB-8 and D101 can except effectively the water-soluble impurities, so the purpose of refining can be achieved.3. Two flavonoids were isolated from the extract of Coreopsis tinctoria by silica gel column chromatography and Polyamide column chromatography. The TLC was used for the qualitative detection of the two flavonoids,and color reaction and the scanning of UV spectrum was used to judge preliminary the structure of these two flavonoids. The result showed that, one of them may be part of Chalcone, and another was belong to Flavonols.And the HPLC was used for the preliminary of separation and analysis of flavonoids in Coreopsis tinctoria.
