Purpose:The research aims at observing the effect on systolic pressure, Ang II,AT1R、p38MAPK and TGF-β1 in DS by electric acupuncture at bilateral "Zu sanli", "Q-uchi" point with ELESA, RT-PCR and WB. It reveals the validity a nd diversity between electric acupuncture at bilateral "Zusanli", "Q-uchi" p oint and non-acupoint,it also analyzes how acupuncture treatment can protec tion of blood vessels and regulate blood pressure through p38 MAPK signal pa thway. It provides scientific demonstration for the mechanism of acupunctur e treatment.Material and method:40 healthy rats,10 of them salt resistant rats as normal group,30 DS were randomly divided into 3 groups(n=10),model group, needling group, non-acupoint group.Everyday the model groupwas bound and fi xed 20 min, not treated with acupuncture; After the needlinggroup was bound and fixed,the needling group was treated by needling "Zusanli", "Q-uchi" poi nt and acupuncture needles for 3mm,then electric acupunctureapparatus was c onnected to conduct dilatational wave stimulation for 20 mi-nutes, the inte nsity depends on vibration of the needle handles, the above operation proce ss was conducted once a day.After the non-acupoint group was bound and fixe d,the needling group was treated by needling self-appointed non-acupoint,an d acupuncture needles for 3mm,then electric acupuncture app-aratus was conn ected to conduct dilatational wave stimulation for 20 minut-es,the intensit y depends on vibration of the needle handles,the above oper-ation process w as conducted once a day.6 times a week,a four-week.At the end of the treatment,taken all the rats’ serum,aortic.AngⅡ were measured by means of ELISA.AT1 R and p38 protein kinase activity were measured by means of WB.the m RNA expression of TGF-β1 were detected by means of fluorogenic quantitative PCR.Results:1. The effect of electric acupuncture of "Zusanli", "Q-uchi" point on the systolic blood pressure of the rats.Compared with the model group, the systolic blood pressure content of the rats in the normal group was lower(P<0.01).After 4 weeks of treatments,ompared with the model group, the systolic blood pressure of the rats in the normal group,needling group and non-acupoint group was obviously reduced(P<0.01);the systolic blood pressure of the needling group was more significant than non-acupoint group(P<0.05).2. The effect of electric acupuncture of "Zusanli", "Q-uchi" point on the AngⅡ content of the rats’ serum. Compared with the model group, the AngⅡ content of the rats in the normal group was lower(P<0.05),the AngⅡ content of the rats in the needling group was obviously reduced(P<0.01),the AngⅡ content of the rats in the non-acupoint group was reduced(P<0.01);the systolic blood pressure of the needling group was more significant than non-acupoint group(P<0.05).3. The effect of electric acupuncture of "Zusanli", "Q-uchi" point on the AT1 R content of the rats’ aortic. Compared with the model group, the AT1 R content of the rats in the normal group was lower(P<0.01),the AT1 R content of the rats in the needling group and non-acupoint group was obviously reduced(P<0.01);the systolic blood pressure of the needling group was more significant than non-acupoint group(P<0.05).4. The effect of electric acupuncture of "Zusanli", "Q-uchi" point on the p38 MAPK content of the rats’ aortic. Compared with the model group, the p38 MAPK content of the rats in the normal group was lower(P<0.01),the p38 MAPK content of the rats in the needling group and non-acupoint group was obviously reduced(P<0.01); the systolic blood pressure of the needling group was more significant than non-acupoint group(P<0.05).5. The effect of electric acupuncture of "Zusanli", "Q-uchi" point on the expression of TGF-β1 m RNA of the rats’ aortic Compared with the model group, the expression of the TGF-β1 m RNA content of the rats in the normal group was obviously lower(P<0.01),the expression of the TGF-β1 m RNA content of the rats in the needling group and non-acupoint group was obviously reduced(P<0.01);the systolic blood pressure of the needling group was more significant than non-acupoint group(P<0.01).Conclusion:1.Electric acupuncture of "Zusanli", "Q-uchi" point has the effects of loweri the systolic blood pressure of the rats. 2.There were diffrences in the effects between Electric acupuncture of "Zusanli", "Q-uchi" point and non-acupoint that used in this experiment. 3.The decompression mechanism of could berelated with the p38 MAPK signal pathway.
