Purpose:This experiment observes the effect of electric acupuncture of tsusanli and quchi acupuncture points on P-selectin,E-selectin in rats serum and the expression of m RNA of the ICAM-1, VCAM-1 and MCP-1 in the thoracic aorta vascular endothelial of prehypertension rats.Discuss abnormal secretion of cell adhesion molecules and chemotactic factor is closely related to occurrence and development of prehypertension,and tries to discusses the effects and action mechanism of acupuncture treatment for prehypertension.Interpret the curative effect of acupuncture treatment of prehypertension and the protection mechanism of the blood vessels.Material and method:7-week-old salt-resistant rats(SS),male,10, weighing 110g-130 g,as normal group; 7-week-old salt-sensitive rats(DS),male,30,weighing 110g-130 g,adopt the method of randomized divided into model group, acupuncture group and non-acupoint group.Adaptive breeding for a week,feed 8%Nacl high-salt diet for a week.During this time,use the non-invasive blood pressure monitor test rat tail artery systolic pressure,after a week,when the rat tail artery systolic blood pressure rose to 120~139/80~89 mm Hg,identified as prehypertension rat model.Stop feeding high-salt diet instead of ordinary diet feeding.Electric acupuncture to be treated at the same time every day:the model group is bound for fixing for 20 minutes and not subjected to acupuncture everyday;the rats in the acupoint group are bound,then tsusanli and quchi acupuncture points are subjected to acupuncture by acupuncture needles for about 3mm,electric acupuncture apparatus is connected to conduct dilatational wave stimulation for 20 minutes,the intension depends on vibration of the needle handles,and the operation is conducted once a day;the rats in the non-acupoint group are bound,then customized non-acupoint parts are subjected to acupuncture by acupuncture needles for about 3mm,the electric acupuncture apparatus is connected to conduct dilatational wave stimulation for 20 minutes,the intension depends on vibration of the needle handles,and the operation is conducted once a day.Continuous acupuncture for 6 days,treatment of 4 weeks,use the non-invasive blood pressure monitor test rat tail artery systolic pressure once a week. After the treatment is completed,take the organization such as serum, thoracic aorta of the rats in each group are taken. The P-selectin, E-selectin in rats serum are detected by using the ELLSA method;The expression of m RNA of the ICAM-1, VCAM-1 and MCP-1 of the thoracic aorta are detected by using the fluorogenic quantitative PCR method.Results: 1. The effect of electric acupuncture of tsusanli and quchi acupuncture points on prehypertension rat tail artery systolic blood pressure After treatment for 4 weeks, compared with the normal group, the rat tail artery systolic pressure in the model group is obviously rised(P<0.01); compared with the model group, the rat tail artery systolic pressure in the acupoint group and non-acupoint group is obviously reduced(P<0.01). 2. The effect of electric acupuncture of tsusanli and quchi acupuncture points on P-selectin of prehypertension rats. Compared with the model group, the P-selectin of the rats in the normal group is obviously reduced(P<0.01);there is no statistical significance by comparing the acupoint group with the normal group(P>0.05), compared with the model group, the P-selectin in the acupoint group and non-acupoint group is reduced(P<0.05). 3. The effect of electric acupuncture of tsusanli and quchi acupuncture points on E-selectin of prehypertension rats. Compared with the model group,the E-selectin of the rats in the normal group is obviously reduced(P<0.01);there is no statistical significance by comparing the acupoint group with the normal group(P>0.05),compared with the model group, the P-selectin in the acupoint group and non-acupoint group is reduced(P<0.05). 4. The effect of electric acupuncture of tsusanli and quchi acupuncture points on the expression of the ICAM-1 gene on the thoracic aorta of prehypertension rats. Compared with the model group, the expression of the ICAM-1 m RNA content of the rats in the normal group is obviously reduced(P<0.01);compared with the model group, the expression of ICAM-1 m RNA content of the acupoint group is obviously reduced(P<0.01);there is no statistical significance by comparing the acupoint group with the normal group(P>0.05); there is no statistical significance by comparing the model group with the non-acupoint group(P>0.05). 5. The effect of electric acupuncture of tsusanli and quchi acupuncture points on the expression of the VCAM-1 gene on the thoracic aorta of prehypertension rats. Compared with the model group, the expression of the VCAM-1 m RNA content of the rats in the normal group is obviously reduced(P<0.01);compared with the model group,the expression of VCAM-1 m RNA content of the acupoint group is obviously reduced(P<0.01);there is no statistical significance by comparing the acupoint group with the normal group(P>0.05); there is no statistical significance by comparing the model group with the non-acupoint group(P>0.05). 6. The effect of electric acupuncture of tsusanli and quchi acupuncture points on the expression of the MCP-1 gene on the thoracic aorta of prehypertension rats. Compared with the model group,the expression of the MCP-1 m RNA content of the rats in the normal group is obviously reduced(P<0.01);there is no statistical significance by comparing the acupoint group with the normal group(P>0.05), compared with the model group, the expression of MCP-1 m RNA content of the acupoint group and non-acupoint group is obviously reduced(P<0.01).Conclusion: 1.Electric acupuncture of tsusanli and quchi acupuncture points has the effects of adjusting blood pressure. 2.The abnormal secretion of cell adhesion molecules and chemotactic factor is closely related to occurrence and development of prehypertension. 3.Electric acupuncture of tsusanli and quchi acupuncture points has the effects of adjusting the abnormal secretion of cell adhesion molecules and chemotactic factor of prehypertension rats,which have an important effect on the protection mechanism of the blood vessels.
